Objective: To investigate the anti-proliferation and inhibiting fiber synthesis of ethanol extract from boschniakia rossica (BR) for Hepatic stellate cell -T6(HSC-T6)Methods: The studv object is HSC-T6 in vitro. Using, the MTT assavand the cell growth curve, the inhibition of BR and interferon for HSC-T6 celTs gro\\th was examined and using the invert microscope, HE stain and the scanning electron microscope, the morphologic change of cells was observed. The effect of medicine related to the cytotoxicity was detected by the LDH activity method . CIV and HA content in the cell culture liquid were determined by the ELISA method for further exploration of the anti-tlbrosis mechanism of BR and IFN.Results: (1) Detection consequence of using MTT method and growth curve show BR and IFN significantly inhibited the proliferation of the HSC-T6 cells in vitro and the inhibitive effects were dependent on the medicine concentration and length of treatment time.(2) LDH activity of experimental group and activity of the experimental group and the control group do not have a significant difference (P>0.05) by detection of the LDH activity. It showed indirectly the anti-proliferation of BR and IFN for HSL-T6 cell is not caused by the nonspecific cytotoxicity.(3) Observation by light and electron microscope show the cell start to have morphologic change from an irregular shape to a round shape, the volume was reduced significantly after treating for 4-6 hours with the BR and karyopyknosis apoptotic bodies were found after 12 hours.(4) The contents of CIV and HA show there is a significant decrease in the content of CIV and HA between the BR group, the IFN group and the control group. ( P<0.01 ). The effect will be more obvious with the increase of medicine concentration and extension of time.Conclusions: BR and IFN has a significant anti-proliferation and inhibition of synthesis of collagen on HSC-T6 cell in vitro.
