Objective:Study on Herba Houttuyniae extract aerosolized anti asthmatic effect and the effective concentration.Methods:Male BALB/C mice of50were randomly divided into5groups, each group had10mice. The normal control group (group A), model group (group B), the low dose Houttuynia cordata group (group C), extract dose Houttuynia group (group D), high dose group Houttuynia (E group). Group B mice sensitized by intraperitoneal injection with50μg of OVA and1mg aluminum hydroxide in200ul of0.9%sterile saline on the day1st and day7th. Then were challenged with aerosolized OVA (1.0%15ml) for20min on the day14th, day21st, day22nd and day23rd. the mouse In group C, group D, group E will be given inhalations of Houttuynia1mg/ml,10mg/ml,100mg/ml15ml for20min before1hours challenge. BALF and peripheral blood were collected on the day24th, and examined the quantity of eosinophils, neutrophils, mononuclear cells, lymphocytes and macrophage by Diff-quik stain, ELISA examined the level of BALF, the infection circumstance of lung by Hematoxylin-eosin and congo-red stainResults:1.The cell changes in BALF:group C, group B compared with group A, the volume of eosinophils, lymphocytes, neutrophils in BALF increased significantly; IL-4, IL-5, IL-6increased significantly (P<0.05); group D, group E compared with group B, eosinophils, lymphocytes, neutrophils, significant decrease in the number (P<0.05); IL-4, IL-5, IL-6decreased significantly (P<0.05).2. The cell changes in peripheral blood:Group B compared with group A, eosinophil count increased significantly (P<0.05); C group, D group, E group compared with B group, eosinophils significantly reduced (P<0.01); group D, group E compared with group C, eosinophils were significantly reduced (P<0.01); in each experimental group neutrophils, lymphocytes, monocytes, no obvious change.3. histological observation showed Houttuynia extract dose group, high dose group significantly reduced airway and interstitial pulmonary pathological changes.Conclusion:From the results of this study, it is suggested that Houttuynia extract dose (10/ml), high dose (100/ml) can effectively prevent and inhibit the OVA-induced bronchial asthma in mice...
