Objective:The research focuses by establishing diethyl nitrosamines (DEN) induce rat liver cancer model, dynamic observation MAPK signalling pathways of regulatory factors position regularly problem,and expounds the origin and development of tumor mechanism.Methods:136Health Wistar rats were divided into two groups:16normal control group(sterilization water),120experimental group (DEN).The experimental group drink99μg/ml DEN freely,a total of22weeks.In all experiment,normal control group drink sterilization water freely.The experimental group rats were executed respectively in5、8、10、12、14、16、18、20weeks each only10-15rats.The normal control group rats were executed respectively in the regular time in5to20weeks.And in different regular time, p38、p-p38、ERK1/2、p-ERK1/2、JNK、p-JNK protein of Liver tissue and tumor tissue were detected by Western Blot. p38、p-p38、ERK1/2、p-ERK1/2、JNK、p-JNK of mRNA by RT-PCR.Results:In rat hepatoma model development, protein of p-p38、p-ERK1/2、p-JNK and its mRNA expression were significantly increased with increasing treatment time, but,no significant changes of p38, ERK, of JNK protein and its mRNA expression.Conclusion:Protein of p38、p-ERK1/2、p-JNK and mRNA expression rise may induce the occurrence and development of the rat liver cancer.
