Objective: To establish an objective animal experimental model for the experimentalstudy of clinical bone defect, detect the BMP-2activity in bone defect area, and observethe early healing situation of rabbit radial fracture. Methods: The experimental objects,72New Zealand male rabbits (weight/each:2.8±0.2kg), were randomly divided into A, B,C, D four experimental groups (18/each), and there were0.5cm,1.5cm,2.0cm, and3.0cm bone defect models ablished respectively in the middle of the rabbit left radius. Bythe time of modeling, retained all the bone segments and took a small amount of boneadjacent tissues as E specimens of control group. Modeling time was recorded as0week,and six animals of each group were put to death in1W,3W,4W later. Then took the bonedefect surrounding tissues, checked BMP-2content by Western-blot Detection, andobserved the bone healing situation at different time. Results: BMP-2compositionshowed a significant increase in secretory volume of each group’s bone defect area tissuesafter modeling one week, compared with E specimens of control group. BMP-2composition secretion capacity reached its peak in the third week and BMP-2contentreduced apparently in the fourth than before. At the same time, BMP-2content decreasedwith the addition of bone defect length. There was also a correlation between bone healingand time with the bone defect length. Conclusion: This study has successfully establisheda comparable rabbit radial bone defect model. These objective animal models, which arethe closest to clinical reality, provide the theoretical basis of animal experiments to explorethe mechanism of human fracture healing.
