Objective:Qi Deficiency and Blood Stasis Syndrome is the common syndrome of many clinical disease which is blood stagnancy based on Qi deficiency in TCM, especially in the cardiac and cerebrovascular disease in the elderly. It is involved in multisystem which emerges in chronic disease. With the rapidly development of population aging in China, the study on Qi Deficiency and Blood Stasis Syndrome is getting more and more important and popular as well as of great social significanceAt present, it is known that age, overtired, improper diet, overly thrilled can cause the damage of Qi and lead to Qi deficiency according to TCM. When Qi deficiency develops to a certain stage, it must lead to Blood stasis. The pathogenesis of TCM. Western anatomy and Chinese-western combined medicine theories can be adopt in the building of animal model. The formation of Qi deficiency and blood stasis syndrome should have some influence on scavenging oxygen free radicals, the hemodynamics, clotting function, energy material changes biochemical indicator.The method of supplementing Qi and activating blood circulation is the common way of therapy to improve symptoms. The Compound Lumbrical Capsule(CLC) is a Chinese patent drug of Lumbrokinase, which is consist of lumbricus, Astragalus, Ligusticum wallichii. Achyranthes bidentata. It can be used to supplement Qi and activating blood circulation and huayutongluo.The method of making model used in this study discuss a new method to form a whole body blood stasis state which by hungry and purgation. The model would be evaluated by the characterization observation, superoxide dismutase(SOD), hemodynamics. function of blood coagulation. The effect of using CLC would be tested by blood vessel damage situation of Qi deficiency and blood stasis syndrome byendothelin-1(ET-1). Nitric oxide synthase(NOS), the numberof endothelial progenitor cells (EPCs) in peripheral blood.Methods1.Establish a suitable model after comparing hungry+purgation(model group-A) and hungry+purgation+macromolecule iron dextran(model group-B) to meet the research purposes.2.Determinate model of Qi deficiency and blood stasis syndrome according to detect the number of EPCs. 3.Study the effect of CLC for Qi deficiency and blood stasis syndrome by detecting superoxide dismutase. hemodynamics activated partial thromboplastin time (APTT) and prothrombin time (PT).4.Measure NOS. ET-1, EPCs in peripheral blood of Qi deficiency and blood stasis syndrome to evaluate blood vessel damage and the repair effect of using CLCResult1. Successfully established the Qi deficiency and blood stasis syndrome model on rat. Both methods of making model could cause the change of hemodynamics and part of the function of blood coagulation.2. The influence of CLC on EPCs in the two model methods is different. EPCs in model group-A is decreased significantly compare with normal group (P<0.05), but there is no statistical difference between model group-B and normal group(P>0.05).3. The activity of SOD can be significantly increased in CLC groups and part of hemodynamics can be efficiently improved. The PT in high and middle dose group were improved compare to the model group (P<0.05). but the effect on APTT are no significant effects(P>0.05). The low dose group has no effect both for PT and APTT.4. The activity of TNOS and iNOS can be significantly increased in CLC group compare to the model group (P<0.05) as well as ET-1activity.5. The number of EPCs can be significantly increased in high and middle dose groups of CLC (P<0.05), at same time, low dose group have no effect on EPCs(P>0.05).Conclusions:The Qi deficiency and blood stasis syndrome model in rat was successfully established. The abnormal changes in hemodynamics, ET-1in model group point out that the vascular endothelial cells maybe have been damaged. CLC can protect cells against toxicity of oxygen from free radicals through enhance SOD activity. It also can improve blood stasis syndrome through hemodynamics and the part of function of blood coagulation. CLC plays an important role in vasomotor function through enhancing TNOS. iNOS activity, reducing ET-1level and protecting against endothelial hurt. It can play a role in endothelial repair through increase the number of EPCs. Qi deficiency and blood stasis syndrome mechanism may be concerned with vascular endothelial injury. Vascular endothelial repair is one of the effective breakthrough points in the treatments of Qi deficiency and blood stasis syndrome using recipes replenishing Qi and activating blood.
