[Background]The aging population problem is considered to be one of the three global social problems of the21st century. WHO definition of aging is that if a country’s elderly population (over the age of60years) accounted for10%of the total population, or7%population over the age of65years, and that the country has entered the aging stage. The developed countries are the first country entered the aging stage. Since the1970s, there is a difference in the age structure of the world population aging process. As of the end of2006, China’s aging population is more than over100million. By2050, China’s aging population will account for a quarter of the total population.Present study consider that aging and geriatric occurrence is associated. Both of them have the same pathophysiological process, such as the doctrine of free radical damage, immune system damage, neuroendocrine disorders, lipid and carbohydrate metabolism disorders and so on. As aging some geriatric diseases incidence increased significantly, such as atherosclerosis, coronary heart disease, diabetes, hypertension, neurodegenerative and so on. Aging will seriously affect the health of the elderly population. Anti-aging will bring great significance to China’s current development.Since ancient times, there are many mechanisms of aging. Now, the study of aging has entered to the molecular level, such as p16-INK4a has been considered tobe a important biological markers of cell aging. Clearing pl6-INK4a positive cells can delay aging. Cellular senescence involves many complex mechanisms of biological molecules. If the regenerative cells instead of senescent cells, just an cell regeneration and aging to achieve a balanced, anti-aging is likely to be a reality. In1493-1541, the Philippines Auredus Parace firstly proposed cell therapy, which could rejuvenation of the body by entering regenerative cells, since the1950s, there were millions of person receiving cell therapy. As one of cell therapies, that stem cell therapy will offer enough cells to anti-aging. Adipose-derived stem cells is already been used to anti-aging and obtain the good clinical effect. Mesenchymal stem cells has been confirmed that it has the anti-aging effects in animal experimental. Taguichi clinical trials confirmed that endothelial progenitor cells can not only promote angiogenesis of ischemic area of the brain, to maintain the function of vascular endothelial cells, but also promote the growth of neurons. This bone marrow-derived hematopoietic stem cells such as CD34will secretion of vascular endothelial growth factor, insulin-like growth factor and fibroblast growth factor, these factors and endothelial progenitor cells involved in the repair of ischemic tissue function and the growth of neurons. The insulin-like growth factor has the effect of anti-aging, but there is no clinical studies endothelial progenitor cells with anti-aging effect have been reported.Currently antioxidant substances, hormones, immune function and scale are used in anti-aging assessment. The free radical theory consider that of aging cell metabolism generates free radicals which play a very important role in human aging. Superoxide dismutase SOD (superoxide dismutase) is one of the most important antioxidant enzymes. It can scavenge superoxide anion radicals, block the free radical chain reactions, prevent organizations from superoxide anion free base damaging. SOD response to the antioxidant capacity, reply the body’s anti-aging ability.Human growth, development and aging is mainly regulated by the hormone levels. Hormone levels changes in different physiological stages, IFG-1have anti-aging effect, is able to replying the degree of aging of the body. Immunological hypothesis that aging immune system participate in the vertebrate aging process, is a major regulator of the aging process systems, by immunoglobulin IgA, IgQ IgM deciding the function of the immune system with aging, blood IgA, IgG elevated but IgM reduced. Scales, such as the aging self-test scale and the Pittsburgh Sleep rating scale can determine the functional state of the body. Physiological cost index PCI (physiological index of Energy Consumption) also assessed the functional state of the body.[Objective]Study of autologous endothelial progenitor cells in the anti-aging and safety with scale and blood sample.[Methods]From October2011to November2012,4patients undergoing treatment in our department were assigned into2groups. The inclusion criteria for the patients:the age between25and80years, voluntarily cooperate with the treatment and follow-up; Exclude criteria:pregnant women, autoimmune diseases, lactating patients and tumor patients, allergic to penicillin and other febrile diseases, liver and kidney respiratory diseases. The experimental group:1male and45years old,1female75-year-old. The experimental group received the endothelial progenitor cell therapy except conventional treatment. The control group:1male and49-year-old and1female77years old. The control group only received traditional conventional treatment such as Chinese medicine treatment and exercise therapy.[Treatment theraphy]1. Before the experiment The experimental group and the control group must test for blood Routine, blood coagulation index, Urine Routine, liver and kidney function, glucose metabolism indicators, indicators of lipid metabolism, heart failure indicators, inflammatory markers, infectious diseases and so on. In the process, we will exclusion the one who can not conform to the standard.2. Endothelial progenitor cells In the experimental group, on the anterior superior iliac spine, we extract bone marrow50ml and30ml blood of autologous peripheral brachial vein. The control group is not needed for this operation.3. Cell culture and identification Cell culture strict GMP (Good Manufacturing Practice) conditions, density gradient centrifugation of bone marrow mononuclear cells. Bone marrow mononuclear cells were seeded at1x105/cm2fibronectin-coated plastic flasks. EC specific culture medium were cultured to go unless adherent cells incubated for4days.2medium was changed every week,80%confluent cells passaged. A professional company will test expression of telomerase, surface markers, finished test, survival, endotoxin, sterility test, mycoplasma, viruses exogenous factor, retroviral, special human-derived virus, cell tumorigenicity, residual serum detection and so on. It takes about a month to complete all project.4. Cell transplantation Before transplantation, all the cells must pass strict quality control. The amount of cell about2.5×106/kg, was suspended in100ml normal saline of5%autologous serum. Then the cell was injected by radial vein. Adverse reactions should be observed in patients in the process closely. About one week, the second transplant was proceed, the number of transplanted cells is the same as the first time.5. Before the experiment and four months after the experiment, we get the index such as IgM, IgA, IgG, SOD, IFG-1, AAS, PSQI and PCI.[Evaluation of curative effect]1. Superoxide dismutase (SOD) 2. Expected to using SPSS13.0statistical software for statistical analysis of the experimental results.[Result]1. Superoxide dismutase (SOD)Compared with before treatment, the superoxide dismutase of blood is increased*after treatment in the two subjects of experimental group. In two subjects of control group, the superoxide dismutase of after treatment than before treatment, one is reduced, anther one is increased.2. Insulin-like growth factor-1(IGF-1)Compared with before treatment, the blood insulin-like growth factor-1(IGF-1) is increased after treatment in the two subjects of experimental group. In two subjects of control group, the superoxide dismutase of after treatment and before treatment did not change significantly3. Immunoglobulin IgA, IgG, IgMIn two subjects of the experimental group, the blood IgA, IgG reduced, IgM increased after treatment than before treatment. In two subjects of control group, the blood IgA, IgG reduced, IgM did not change significantly.4. Aging self-test Scale rating scale (AAS)In two subjects of the experimental group, compared with before treatment scale score of after treatment is reduced than before treatment. In two subjects of the control group, scale score of after treatment is reduced slightly than before treatment.5. Pittsburgh Sleep Index scale (PSQI)In two subjects of the experimental group, compared with before treatment scale score of after treatment is reduced than before treatment. In two subjects of the control group, scale score of after treatment is also reduced than before treatment.6. physiological index of energy consumption (PCI)In two subjects of the experimental group, compared with before treatment PCI of after treatment is reduced than before treatment. In two subjects of the control group, PCI of after treatment is reduced slightly than before treatment.[Conclusion]Through clinical observation, the autologous endothelial progenitor cells improved the function of the subjects body of the experimental group such as immune system function, anti-oxidation ability, hormone secretion levels, physiological index and aging self-test consumption level. The autologous endothelial progenitor cells is safe, reliable and have no immune rejection and other adverse reactions in the course of treatment and follow-up. Autologous endothelial progenitor cells may become a new research topic of ant-aging.
