Objective:To observe the influence of Ziyin Huoxue Jiedu decoction (ZHJD) on the expression of MCP-1, NF-kBp65of brain tissue in rats after the MCAO model of focal cerebral ischemia, as well as to explore the effect of Ziyin Huoxue Jiedu decoction on the intervention of the inflammatory response after acute cerebral ischemia.Methods:The32healthy adult SD rats were selected. The rats were randomly divided into4groups:the sham-operated, ischemia, ischemia with Ziyin Huoxue Jiedu decoction (ZHJD), ischemia with Argatroban injection(AI), eight rats in each group. The rats in sham-operated group were separated the blood vessels in brain, not to model-processed. The other three groups were made with the MCAO model. The brains were removed after24h. The expression of MCP-1and NF-kBp65were detected by immunohistochemical technique.Results:1.Compared with the sham-operated, the score of neuroethology in model group, ZHJD group and AI group increased on the2h and24h points (P<0.001). Compared with2h point, the score of neuroethology on24h point in sham-operated group and model group didn’t decrease significantly (P>0.05); the score of neuroethology on24h point in ZHJD group and AIgroup decreased significantly (P<0.05).2. Compare with sham-operated group, density mean of MCP-1in model group increased significantly (P<0.05); density mean of MCP-1in ZHJD group and AI goup didn’t increase significantly (P>0.05). Compare with model group, density mean of MCP-1in ZHJD group and AI goup decreased significantly (P<0.05). Compare with AI goup, density mean of MCP-1in ZHJD group didn’t decrease significantly (P>0.05).3. Compare with sham-operated group, density mean of NF-kBp65in model group, ZHJD group and AIgroup all increased significantly (P<O.001); Compare with model group, density mean of NF-kBp65in ZHJD group and AI goup decreased significantly (P<0.001). Compare with AI group, density mean of NF-kBp65in ZHJD group didn.t decrease significantly (P>0.05).Conclusion:The ZHJD can improve cerebral ischemia neurological deficits. This decoction can inhibit the expression of MCP-1, NF-kBp65to reduce the inflammatory response after cerebral ischemia.
