Objective:To investigate the effects of Huazhuo Jiedu Huoxue Tongluo prescription on apoptosis and expression of Bcl-2 and Bax in rats of Cerebral Ischemia Reperfusion Injury, from the perspective of apoptosis related gene reveals its neuroprotectiive mechanism.Methods: We established the mice model of cerebral ischemia reperfusion injury(CIRI) by using Zea-Longa method.Fifty-four healthy male SD mice were randomly divided into six groups: the sham operation group, the model group, the aspirin group, the low dose of Huazhuo Jiedu Huoxue Tongluo prescription(the low dose group for short), the middle dose of Huazhuo Jiedu Huoxue Tongluo prescription(the middle dose group for short) and the high dose of Huazhuo Jiedu Huoxue Tongluo prescription(the high dose group for short), nine mice in each group. All mice were returned to their cages after the operation, ate food and water freely after models were successfully established, and the drugs were given by gastric perfusion the next day. Physiological saline were given to the mice in the sham operation group and the model group. The aspirin group were given the same amount of aspirin solution and the respective dose of Huazhuo Jiedu Huoxue Tongluo prescription were given to the low dose group?the middle group and the high dose group. The drugs were given once a day and for 14 days continuously.After that,the animals were neurologically assessed on a 5 Longa point scale,and then beheaded. Each group of rats' right cerebral hemisphere by HE staining,we observed under the shape variation of ischemic brain tissue, and the expression of Bcl-2 and Bax were detected by The PCR method,and the the neuron apoptosis rate was detected by TUNEL.Results:1 The Neurologic deficits symptoms score of each group: Compared with the sham operation group, the scores of the model group increase obviously(P<0.05); compared with the model group, the scores of the middle dose group? the high dose group and the aspirin group decrease(P<0.05), but the low dose group had no significant difference with the model group(P>0.05); the score of the high dose group has no significant difference with the aspirin group(P>0.05).2 Tunel staining positive cells results show that: compared with sham operation group, the rest of the group cell apoptosis rate were significantly increased(P<0.05); Compared with model group, the treatment group, apoptosis rate declined(P<0.05); Aspirin group and high dose group no significant statistical difference(P>0.05); Compared with the high dose group, the middle dose group no decrease apoptosis rate(P>0.05).3 The Bcl-2 gene positive cells expression results: compared with sham operation group, model group the Bcl-2 gene expression increased significantly(P<0.05); Compared with model group, the treatment group the Bcl-2 gene expression levels were elevated with significant difference(P<0.05); Compared with aspirin group, Chinese medicine groups raise the Bcl-2 expression effect is significant(P<0.05); And compared with the low dose group, the Bcl-2 expression of the middle dose group and high dose group increased significantly(P<0.05).4 Bax gene positive cells expressing the results showed that compared with sham operation group, the rest of the group of Bax gene expression levels are higher in significant difference(P<0.05); Compared with model group, the expression of Bax dosage group was obviously lower(P<0.05); Compared with aspirin group, low dose group Bax expression significantly higher(P<0.05); Compared with the low dose group,the middle dose group and high dose group expressed decreased significantly(P<0.05).Conclusion:1 Huazhuo Jiedu Huoxue Tongluo prescription can significantly reduce the neurologic deficits symptoms score in CIRI model rats, and improve the Results: pathological morphological damage in the ischemic regions, promote the recovery of neural function.2 Huazhuo Jiedu Huoxue Tongluo prescription can increase the expression of Bcl-2,reduce the expression of Bax and TUNEL positive cells after cerebral ischemia reperfusion,which might be the mechanism of neuroprotections.
