Abstract:Objective To investigate the protective effects of polysaccharopeptide (PSP) on the immunosuppression mice.Methods32mice were randomly divided into4groups. The mice in normal control group and model group were orally administrated with saline water,the high dose PSP group(500mg/kg/d) and the low dose PSP group (125mg/kg/d) were received PSP treatment for25days, once a day. On the17th day and21th day, all groups except normal control group were injected with cyclophosphamide (150mg/kg/d, i p) to induce the immunosuppression model. The absolute numbers of WBC, CD3+CD4+T cells, CD3+CD8+T cells and CD3-CD19+cells in peripheral blood were detected by flow cytometer and haemacytometer. The thymus index and spleen index were analysed; The relative expressions of interleukin-2(IL-2), interleukin-4(IL-4), transcription factor T-box-containing protein(T-bet), GATA binding protein3(GATA-3) mRNA in spleen tissues were measured by reverse transcription-polymerase chain reaction(RT-PCR).Results:On the22nd day, compared with the normal control group, the absolute numbers of WBC in peripheral blood were all decreased in the model.group and psp-treatedt group,howevery the absolute numbers of WBC in the low dose PSP group were higher than in the model group. The absolute numbers of CD3+CD4+, CD3+CD8+, CD3-CD19+cells in the model group were lower compared with normal control group and the difference was statistically significant (P<0.05). The down degree of lymphocyte subsets in PSP treated groups were lower than that of the model group, and they had statistical differences. The absolute numbers of CD3+CD4+, CD3+CD8+T cells had not statistically differents between the low-dose PSP group.and the normal control group. On the26th day, the absolute numbers of WBC were restored in the PSP treated group. The low dose of PSP significantly improved the total numbers of T-lymphocyte subsets and B-lymphocytes, CD3+CD4+, CD3+CD8+T lymphocytes absolute counts significantly increased,compared with normal control group and the model group (P<0.05). The absolute numbers of CD3-CD19+cells in in the model groups were lower than those of in PSP treated group.But the absolute numbers of CD3+CD4+T cells in the model group restored to the normal level. PSP had no significant effect on the weight and indexes of thymus and spleen in each group; There were no obvious differences in relative expressions of IL-2mRNA and T-betmRNA. However the IL-4and GATA-3mRNA relative expression levels in the model group were significantly higher than the normal control group and PSP low dose group (P<0.05); There were no statistical differences between the normal control group and PSP low dose group in the ratio of IL-2/IL-4and T-bet/GATA-3, but in the PSP low dose group, they were significantly higher than the model group (P <0.05).Conclusion:PSP can extraordinarily improve the numbers of WBC, T-lymphocyte subsets, B-lymphocytes of immunosuppressive mice and balance Thl/Th2response.
