| Apium graveolens L.(celery), is a biennial plant from the family of Umbelliferae, can be found throughout Europe, the Mediterranean, and part of Asia. The seeds of celery contain a variety of volatile constituents which have long been employed as a popular aromatic herb, spice and herbal medicines. Preresearch showed that celery seeds extracts (CSE) have good anti-inflammatory effects, lower serum uric acid levels and have some protective effects on the kidney damage caused by treatment of gouty arthritis. Till now, Toxicology haven’t give any apparent toxic and side-effects on celery seeds extracts. That’s to say, celery seeds can be developed as a new anti-gout drug, which has a bright future.To identify the effects for celery seeds to treat or protect gout and diseases associated, My study carried out several studies as follows.1. Fundamental Research(1) Fingerprint analyze with celery seeds(Green celery seeds, Yellow celery seeds. Yellow heart celery seeds and western celery seeds) from different regions and time. We established analysis method and predict stability of celery seeds in different time and regions from fingerprint analyze results. Early study and patent revealed that the active constituents in celery seeds are flavonoids. We investigated total flavonoids in different celery seeds. From the results we come to conclusion that green celery seeds and yellow heart celery seeds are the best choice for our research. This study provides fundamental datas for the establishment of Quality standard of celery seeds.(2) To identify the major chemical constituents of celery seed extracts. We carried out LC-DAD-MS/MS by using the established analysis method.15compounds are identified including5phthalides and10flavonoids. They are Luteolin7-O-apiosylglucoside, Luteolin7-O-glucoside, Apiin,3’-methoxyapiin, Chrysoeriol7-O-β-D-glucoside, Luteolin, Quercetin, Apigenin, Chrysoeriol, Sudachitin, Senkyunolide F, Senkyunolide G, Sedanenolide, Sedanolide,3-n-butylphthalide.(3) Isolated7compounds from CSE selectively and identified by spectrum analytical technique including HRMS,1Hã€13C-NMR. They were Butylphthalide, Sedanenolide, Sedanolide, Senkyunolide G, Senkyunolide F, Luteolin7-O-β-D- Glu, Chrysoeriol7-O-β-D-Glu. And Senkyunolide G, Senkyunolide F are first time isolated from celery seed.(4) Study on a further XO inhibitory activity of5isolated compounds, We found that Luteolin7-O-β-D-Glu have good activity with inhibition of74.23%at50μg/mL. Butylphthalide, Sedanenolide and Sedanolide showed great XO inhibition without concentration dependent, which means that they are the active parts.(5) Pharmacodynamics study of Butylphthalide, Sedanenolide, Sedanolide. The results showed that they have obvious lower serum uric acid and analgesic activities, which proved phthalides are anti-gout active constituents.2. New drug study(1) Optimize the extract and isolated technology of active parts in celery seeds with the index of flavonoids contents.(2) Set up assaying quality standards of celery seeds crud drugs and study preliminary stability assessment of crude drugs.(3) Pharmacodynamics study of crude drug of CSE. The results showed that they have obvious lower serum uric acid and analgesic activities.(4) Technical study of preparation of Pharmaceutics and conform capsule prescription and set up a Quality control method which is applied to celery seeds.In a word, Our experiment datas provide us lots of evidence in anti-gout activities and the results draw a conclusion that celery seed extracts can be developed as a new antig-out drug. |
