Objectives: To study the effect of combined anticoagulants and agonists onproliferation of adipose derived stromal cells in auto-platelet rich plasma gel stentMethods:ADSCs,implanted into auto-platelet rich plasma (PRP) gel stent usingdifferent anticoagulants (EDTA,heparin) and agonists(thrombin, collagen-Ⅰ),weredivied into EDTA and thrombin group, EDTA and collagen-Ⅰgroup, heparin andthrombin group, heparin and collagen-Ⅰgroup,and control group.ADSCs werecounted everyday for7days and cell growth curve was plotted. Survival rate andproliferation activity of ADSCs were analyzed by methyl thiazolyl tetrazolium(MTT) assay on day2、3、4and6. Growth factor TGF-β1and collagen-Ⅱweredetected by ELISA. Expression level of sox-9in ADSCs wasmeasured byRT-PCR.Results: The platelet counts in the whole blood had a mean value of(181.75±41.44)×10~9/L, and platelet counts in PRP with EDTA had a mean valueof (1040.01±159.72)×10~9/L (5.7baseline) while with Heparin (795.01±140.69)×10~9/L (4.4baseline).The proliferation activity of ADSCs was significantly higherin EDTA and thrombin group,EDTA and collagen-Ⅰgroup, heparin and thrombingroup, heparin and collagen-Ⅰ group,especially in EDTA and thrombingroup,than in control group (P <0.05). Auto-platelet rich plasma gel couldsignificantly promote the release of TGF-β1, synthesis of collagen-Ⅱ as well asexpression of sox-9, especially in EDTA and thrombin group (P <0.05). Conclusions: The effect of auto-platelet rich plasma gel is the best onproliferation of ADSCs in EDTA and thrombin group.
