Objective:The study is to explore the effect of SVF-GEL-assisted lipotransfer through SVF-GEL combines with the adipose granule,and compared with PRP/PRF auxiliary fat grafting,and eventually screens out the optimal portfolio that improves the early vascularization and the survival rate,thus,provides the experimental basis for the clinical practice.Methods:Use syringe to aspirate the adipose tissue,respectively injecte the adipose granule or their compounds into the subcutaneous of both back sides of the BALB/c nude mouse with a blunt needle.The experiment was divided into 5 groups,with fiften nude mouse each group,including AG+NS group?AG+PRP group?AG+PRF group?AG+SVF-GEL group ? SVF-GEL+NS group.Observing the general form and adipose volume of the adipose granule or its compounds of fat grafts at postoperative 1week?2week?4weed?8week?12week.The sample was sliced and paraffin embedding immunofluorescence,then each cell was observed and counted on the microvascular density.Statistical analysis was carried out on the survival rate of fat grafting and microvascular density on the comparison between the different groups.Res?lts:1?Observation of general form:after 1week,the color of five groups of grafts was similar to normal adipose tissue,and the form of normal fat was slightly loose;None of the five groups saw fat liquefaction and necrosis;thereinto,the AG+SVF-GEL group occurs some small vascular clusters on the surface.At 2week,the five groups of fat grafts were similar to the normal adipose tissue,with no significant difference between the groups;Each group showed some fat liquefaction,no necrosis and cyst formation;Expected the AG+NS group,others were all visible in small vascular clusters,howerer SVF-GEL+NS group was the most intense.After 4W,the colour and lustre of five groups are similar to the normal adipose tissue,no significant difference;The five groups still showed a small amount of lipid liquefaction,which was less liquefaction than 2week,and had no necrosis,cyst,hard junction,and calcification;all can see the small blood vessels cluster formation,AG+NS group?AG+PRP group?AG+PRF group are no difference between the small blood vessels cluster area,is least in 5 groups;AG+SVF-GEL group is second,SVF-GEL+NS group is the most densest.At 8week,the colour and lustre of the fat grafts each group were similar to normal adipose tissue;Fat liquefaction in five groups was rare,and the AG + PRP group occuered an example of necrosis,which was not seen in cyst,hard junction,and calcification;The small vessels cluster in each group were smaller than before.12 W,the five groups of grafts were formed with normal adipose tissue,without liquefaction,necrosis,cyst,hard junction and calcification.2?Fat volume retention rate: at 1 week,2 week,4week after fat grafting,the volume survival rate of AG+NS group and AG+PRP group,there were no obvious statistical significance difference(P>0.05),and compared with AG+PRF group,AG + SVF-GEL,SVF-GEL + NS group,there were statistically significant difference(P<0.05);AG +SVF-GEL has the highest volume retention rate,and SVF-GEL group has the lowest volume retention rate;At 4week,the AG+PRP group and the AG+PRF group were not statistically significant(P>0.05).At the 8week,the difference between the AG+NS group and AG+PRP group,AG+PRF group,AVF-GEL+NS group was not statistically significant(P>0.05).At the 12 week after fat grafting,the difference between the AG+PRP group and the AG+PRF group was not statistically significant(P>0.05);The difference between the AG+NS group and svf-gel+NS group was not statistically significant(P>0.05),which was different from the AG+PRP group,AG+PRF group,AG+svf-gel group(P<0.05).3?paraffin embedding and immunofluorescence microsection:the microvascular density increased steadily after the transplant,and the peak was at 4 week.At 2week,the difference among the AG+SVF-GEL group,the AG+PRF group and the SVF-GEL+NS group was not statistically significant(P>0.05),which was better than the AG + NS group,AG + PRP group.At 4week the adipose tissue of the AG + svf-gel group was the highest,significantly different than the other four groups(P < 0.05).At 8week,the difference between AG+SVF-GEL and AG+NS was statistically significant(P<0.05).There was no significant statistical significance(P > 0.05)in the vascularization between the groups at 12 week.Conclusions:SVF-GEL-assisted lipotransfer accelerates the fat tissue remodeling early by promoting early fat tissue vascularization and into fat,so as to improve the survival rate of the fat grafts.
