Objective:The present study was designed to investigate the protective effect of curcumin intervention on renal injury in exhausted exercise rats and its mechanisms.Methods:This study used healthy56male SD rats,210±10g body weight, they were selected and randomly divided into7groups according to their weightnormal control group, exhaused model group, exhaused+western medicineof losartan group(20mg/kg),exhaused+low-dose curcumin group(100mg/kg), exhaused+middle-dose curcumin group(200mg/kg) exhaused+hyper-dose curcumin group(300mg/kg) and curcumincontrol group(200mg/kg),each with eight.Normal control group untake any exercise,common breed;all exercise groups held to exhausted exercise; exhaused+curcumin groups treated with different dose of curcumin (100mg/kg,200mg/kg,300mg/kg); exhaused+western medicine of losartan group treated with losartan (20mg/kg);curcumin group treated with200mg/kg of curcumin.After four weeks,accurate records of the time immediately after exhaustive exercise.HE staining of histopathological changes of renal was detected with optical microscope,using bio-chemical analyzer,contents of serum BUN,Cr,SOD,MDA were detected,cytokines TNF-α,IL-1β,IL-18,TGF-βcontent in kidney and blood was detected by double-antibody sandwich enzyme-linked immunosorbent assay (ELISA), Caspase-3is measured quantitatively using colorimetric assay kit.Results:(1)Curcumin can significantly extend the exhausted exercise rats exhaustive exercise duration, the difference was statistically significant compared with exercise model group (p<0.05)(2) Compared with exercise model group,treatment with curcumin resulted in significant reduction in serum and tissue level of MDA,BUN,Cr (p<0.05),the level of SOD increased signifieantly (p<0.05) comparing the BUN,Cr,MDA and SOD of blood-serum and renal tissue among groups of different doses of curcumin,middle-dose have difference and its effect to BUN,Cr,MDA and SOD surpass to other two groups (p<0.05);there is no difference between middle-dose curcumin group and western medicine group (p>0.05)(3)Compared with exercise model group, both systemic as well as blood levels of TNF-α,IL-1β,IL-18and TGF-β and caspase-3decrease significantly in exercise+curcumin group rats (p<0.05) comparing the TNF-α, IL-1β,IL-18,TGF-(3and caspase-3of blood-serum and renal tissue among groups of different doses of curcumin,middle-dose have difference and its effect to TNF-α,IL-1β,IL-18,TGF-pand caspase-3surpass to other two groups (p<0.05);there is no difference between middle-dose curcumin group and western medicine group (p>0.05)(4) By optical microscopy observation of renal tissue,we found that use of curcumin the rat kidney tissue ultra structural,pathological damage obviously improved.Conclusion:(1)Curcumin can significantly prolond exhaustive exercise time.(2)Curcumin can alleviate renal injury by increasing the antioxidant enzyme activities and clear the role of free radicals.(3)Curcumin can improve kidney damage by inhibiting pro inflammatory cytokines TNF-α, IL-1β, IL-18, TGF-β generation and caspase-3activation.(4)Analyzed the change of oxygen free radicals and pro-inflammatory cytokines,combined with the morphological observation, middle dose of curcumin (200mg/kg) had more obvious renal protective effect than the other doses.
