Detection And Significance Of Serum Interferon-gamma And Interleukin-2and Interleukin-10in The Patients With Chronic Hepatitis B Overlapping Acute Hepatitis E

Object Detecting the levels of three serum cytokines(IFN-gamma, IL-2and IL-10),the main liver function and HBVDNA in the patients with chronic hepatitis Boverlapping acute hepatitis E to discuss the aggravating mechanism of HBVsuperinfected HEV and to discuss the relationship between the three serumcytokines(IFN-gamma、IL-2、IL-10) and replication of HBVDNA.Methods The patients’ serum level of IFN-gamma, IL-2and IL-10of chronichepatitis B overlapping acute hepatitis E (overlapping group), acute hepatitis E (AHEgroup), chronic hepatitis B (CHB group) and the healthy check-up(control group)were determined by using Enzyme-linked Immunosorbent Assay test (ELISA). Usingreal-time tracking fluorescence PCR to determine HBVDNA of overlapping groupand CHB group, and using ELISA to determine the serum markers of CHB group.Themain liver function of overlapping group, AHE group and CHB group weredetermined.Results The IFN-gamma in overlapping group was significantly higher than that inCHB and control groups, while the IL-10in overlapping group was lower than that inCHB and control groups, IL-2in overlapping group was higher than that in CHBgroup, IL-10in CHB is the highest level(P <0.05). The main liver function such asAlanine aminotransferase(ALT), Aspertate aminotransferase（AST）, Total bilirubin（TBIL）in overlapping group increased obviously than that in CHB group, andCholinesterase(CHE), Albumin(ALB), Prothrombin activity(PTA) in overlappinggroup decreased obviously than that in CHB group (P <0.05). The negative rate ofHBeAg and HBVDNA in overlapping group were significantly higher than those in CHB group（p=0.00）. The serum level of IFN-gamma and IL-2in overlapping groupwhich HBVDNA was negative were significantly higher than positive one,and theserum level of IL-10is lower than HBVDNA positive one（p﹤0.01）.Conclusions IFN-gamma, IL-2and IL-10were involved in the immunologic injuryprocess that occurred for HBV overlapping HEV. Detecting the serum levels of IFN-gamma, IL-2and IL-10can be used as index of hepatic cell damage degree of HBVoverlapping HEV. IFN-gamma and IL-2may have some effection in HBV replication.
