The Study On The Role Of Dalteparin Sodium On VEGFR And HIF-1α In Human Lung Cancer Cell Line A549

Objective: Different concentrations of heparin on human lung cancer cell line A549VEGF, VEGFR, and the role of HIF-1ɑstudy of the mechanism of inhibition of tumorcell proliferationMethod:1. In cultured human lung cancer cell line A549with different concentrations ofheparin effect on lung cancer cell line A549for24hours,48hours.2. SYBR Green I real-time fluorescence PCR detection of A549cells, VEGF, VEGFR,and HIF-1ɑ of mRNA expression, and different concentration of heparin treatmentA549cells after24h and48h VEGF, VEGFR, and HIF-1ɑ the expression of mRNA.3. Immunohistochemical assay with A549cells, VEGF, VEGFR, and HIF-1ɑ proteinexpression, and different concentration of heparin treatment A549cells after24h and48h VEGF, VEGFR, and HIF-1ɑ expression of the protein.Results:1. VEGF, VEGFR, and HIF-1ɑare expressed in lung cancer A549cells, differentconcentrations (5IU/ml,15IU/ml,50IU/ml,150IU/ml) of the heparin effect on A549cells up to24hours and48hours later, in addition to5IU/ml24h, groups, VEGF,VEGFR mRNA and protein expression compared with the control group was notsignificant (P>0.05), the rest of the group of VEGF, VEGFR, and HIF-1ɑ proteinexpression compared with control group were significantly decreased VEGF, VEGFR,and the expression of HIF-1ɑwith the drug concentration increased and decreasedgradually with time, a clear dose-and time-dependent, the difference was significant.(P <0.05)2. Lung cancer A549cells, VEGFR, HIF-1ɑthe mRNA and protein expression ofVEGF mRNA and protein expression was significantly correlated (P <0.05).Conclusion:1. Lung cancer line A549cells, there is an autocrine mechanism for VEGF/VEGFR; 2. Dalteparin sodium can be reduced VEGF、VEGFR expression in cells, destructionof VEGF/VEGFR autocrine loop, resulting in inhibition of tumor angiogenesis inlung adenocarcinoma A549cells, and the added value of the biological role;3. Dalteparin sodium can be reduced HIF-1ɑ mRNA and protein levels, indirectlyinhibit the expression of VEGF.
