Objective:This study was designed to primarily investigate the inhibitoryeffects and phase growth arrest of sodium phenylbutyrate(PB) on lung cancercell line A549 and YTMLC in order to acquire more information about PB'functions on the treatment of non-small cell lung cancer.Methods:MTT(Methy thiazolyl tetrazolium) assay was used to evaluatethe inhitory effects of PB in different concentration and time. Also MTT wasused to examine the growth inhibition of A549 and YTMLC cells treated by thecombination of PB with Gemzar and Cisplatin. Alteration of morphology of PBinduced on A549 and YTMLC cell line in vitro were observed by invertmicroscope.FCM(the fluorescence flow cytomtry) DNA assay were applied todetect the changes of apoptotic rate and cell proliferation and alteration of cellcycle phase.Results: PB could inhibit the growth of A549 and YTMLC and the inhibitoryeffects showed time-and dose-dependent pattern.There was no differencebetween combination of PB group and the control group without PB.Morphological changes were observed by invert microscope. FCM assayindicated that most of the cells were arrested in G0/G1 phase and the apoptoticpeak appeared. During the prolong of incubation time, the apoptosis rate wasincreased at 72 hours.Conlusion: PB can inhibit the growth of the lung cancer cell line A549 andYTMLC cells in vitro. The inhibitory effect of sodium phenylbutyrate on A549and YTMLC may through the increasing of G0/G1 fraction with a concomitantdrop in the S-phase and apoptosis induction.
