Objective:To investigate the expression levels of Treg and Th17in myelodysplasticsyndrome,then explan the relationship between the Treg and Th17with MDS.Methods:Seventeen untreated MDS patients and ten controls were enrolled in thisstudy.The expression levels of Treg and Th17were examined with the flowcytometric assay and the level of Foxp3mRNA and RORc mRNA were measuredwith RT-PCR. The expression levels of IL-10and IL-17were examined withELISA。Results:1.Compared with controls, the ratios of Tregs in CD4+T cells are higher in MDSpatients,especially in high-risk MDS patients【(2.68±0.48%)vs(5.42±1.18%)vs(9.13±1.65%)】(p<0.05). The same trend also occurred in the the expression ofFoxp3mRNA and the expression of IL-10【(0.20±0.18) vs (0.28±0.52) vs(0.69±0.06)】and【(3.52±1.89)vs(6.14±0.43)vs(9.56±0.94)】(p<0.05).2. The expression levels of Th17in CD4+T cells and RORc mRNA in low-riskMDS patients are significiantly higher than that of high-risk MDS patients andcontrols 【(6.24±1.93%) vs(13.84±3.79%)vs (8.67±1.01%)】(p<0.05).and【(0.20±0.02%)vs(0.57±0.08%)vs(0.29±0.06%)】(p<0.05).The expression of IL-17are highly rise in low-risk MDS,but the rest of the differences between the two groupswas not apparent【(8.05±0.37)vs(16.71±1.36)vs(9.82±1.09)】.3.The ratio of Treg/Th17is notable increased in the high-risk group(1.08±0.26)(p<0.05),but there was not statistically different between high-risk MDS patients andcontrol【(0.42±0.16%)vs(0.38±0.08%)】(p>0.05).Conclusion:The imbalance of Treg/Th17plays an important role in the pathogenesis ofMDS.
