Objective:To study the genotype and gene distribution of HSP70-1(+190G/C), HSP70-2(+1267A/C) and HSP70-hom (+2437T/C) in Jiangxi Han patients with CHD andnormal population and to investigate the association between these three genespolymorphism and the risk of CHD.Research Object:All subjects were selected from cardiovascular department of the secondaffiliated hospital of Nanchang university between Jun.2009and Dec.2010anddivided into CHD group and nomal control group.There were237case in the CHDgroup(malel52,female85,median age63.35±7.77),and203case in the control group(male122,female81,median age62.72±9.18).They were all Jiangxi Han people andhad no blood relstionship.These circumstances must be eliminated,such as infection,immuno-inflammatery disease,malignant tumor,liver and renal dysfunctions andusing immunosuppress-sants.The diagnostic standard for CHD patients meet thediagnose and treatment standard of Chinese medical angiocardiopathy association,while angina pectoris patients were angiographically demonstrated (at least onesignificant stenotic lesion of the major coronary arteries stenosis,in which the luminaldiameter was less than50%of the adjacent unaffected site). Meeting the followinginclusion standard:those without pectoralgia histroy,with normal ECG and withoutcoronary artery calcification found in chest CT;and/or those with pectoralgia but nocoronary artery stenosis was founded by coronary angiography.Methods:Ask the general data about the subjects,such as age,weight,height,smokingrate,family history and so on. Subjects after fasting more than ten hours were takenvenous blood sampling. Triglyceride (TG), low-density lipoprotein (LDL),high-desitylipoprotein (HDL) and fasting blood sugar were detected by standard blood enzymecolorimetric in our hospital.Another5ml venous blood were taken to extractleukocyte genomic DNA from by ways provided by the blood DNA extraction kit. The polymorphism of heat-shock protein70-1gene+190G/C site, the heat-shockprotein70-2gene+1267A/G site,the heat-shock protein70-hom gene+2437T/C sitewere all detected by polymerase chain reaction-restriction fragment lengthpolymorphism(PCR-RFLP)method. The data was analysised by Hardy-Weinbergalignment inspection,Chi-square,t-test and Logistic Regression in SPSS19.0.Result:1. The HSP70-1gene+190G/C polymorphism, the HSP70-2gene+1267A/Gpolymorphism and the HSP70-hom gene+2437T/C polymorphism were found inJiangxi Han population.2. The allelic distribution of the HSP70-1gene、the HSP70-2gene and theHSP70-hom gene in both CHD and control groups were in the Hardy-Weinbergequilibrium.3. The prevalence of HSP70-1gene C allele and(GC+CC)genotype in CHDgroup was statistically significantly higher than in control subjects (χ~2=13.781,P<0.005and χ~2=12.885,P<0.005).4. The prevalence of HSP70-2gene G allele and(AG+GG)genotype in CHDgroup was statistically significantly higher than in control subjects(χ~2=7.577,0.01<P<0.025'χ~2=7.023,0.005<P<0.01).5. The prevalence of HSP70-hom gene T allele and(TC+CC)genotype in CHDgroup was not statistically significantly higher than in control subjects(χ~2=0.343,0.75<P<0.9'χ~2=0.139,0.5<P<0.75).6. After multiple Logistic regressions,we can know MBI,smoking rate,hypertension history, TG, LDL,fasting blood sugar,HSP70-1(GC+CC) and HSP70-2(AG+GG) genetype may the independent risk factors of CHD,but HSP70-hom(TC+CC) genetype was not.Conclutions:1. The HSP70-1+90G/C gene polymorphism was associated with the risk ofCHD.The (GC+CC) genotype and the C allele may the independent risk factors ofCHD.2. The HSP70-2+1267A/G gene polymorphism was associated with the risk ofCHD.The (AG+GG) genotype and G allele may the independent risk factors CHD. 3. The HSP70-hom gene+2437T/C polymorphism was not associated with therisk of CHD.
