Objective: To investigate the effect of MDR and its possiblemechanism about combined application with curcumin and verapamil onhuman gastric carcinoma SGC7901/ADR cells.Methods: Drug sensitivity tests were analyzed by MTT method,RT-PCR was used to determine the changes of the mRNA of MDR1gene,Western blot and Immunofluorescence were used to test the changesof the expression level and the expression site of the P-gp.Results:Using either non-cytotoxic dose25umol/L curcumin or10mg/L verapamil can make SGC7901/ADR cells more sensitive todoxorubicin hydrochloride drug, at the same time can decrease SGC7901/ADR expression level of MDR1and P-gp glycoprotein expression, thecombination use has more significant effect.Conclusion:Curcumin and verapamil have coordinative reverse effecton SGC7901/ADR cells,their possible mechanism might be related toinhibition of MDR1gene and P-gp protein overexpression.
