Objective Use L-arginine join people liver cell LO2cultures,Toexplore the restorability of L-arginine on expression of human FⅧ genein human normal liver cells LO2.Methods LO2were divided into experimental groups and controlgroups, which received L-arginine (final concentration of10mmol/L) fordifferent time periods (24h,36h,48h,60h) and no treatment, respectively.Cells were harvested for detecting human FⅧ mRNA by RT-PCR.Theculture medium was collected for determining the clotting activity ofhuman FⅧ (FⅧ:C).When treated with L-arginine for24h and48h, theexpressions of FⅧ in cells were detected by Western blotting and laserscanning confocal microscopy.Results After the cells were treated with L-arginine for differenttimes(36,48and60h), the expression of FⅧ at mRNA in LO2wererestored, while the control group did not appear human FⅧ mRNAtranscription. FⅧ:C had no diversity in L-arginine group and controlgroup (P>0.05).FⅧ was detected in the cells which were treated with L-arginine for48h by Western blotting and laser scanning confocalmicroscopy, but the protein could not be detected in the control group.Conclusion L-arginine can restore the expression of human FVIIIgene in LO2.
