| Objective: To explore the expression of PRMT1-ADMA-DDAH1axisin kidney tissue of diabetic rats, and the effects of Telmisartan on this axisand the function of kidney.Methods:30male SD rats were respectively assigned to normalcontrol group (NC, n=10), diabetes group (DM, n=10) and Telmisartangroup (TM, n=10). Type2diabetes mellitus models were established in ratsby injecting streptozotocin (30mg/Kg) intraperitoneally after feeding withthe diets enriched with high-lipid and sucrose for4weeks. Telmisartan(TM) group was feeding with the diets enriched with high-lipid and sucroseand telmisartan (10mg/Kg).8weeks later, the content of ADMA in kidneytissue was measured by ELISA, the expression of PRMT1was observed byimmunohistochemistric and western blot. The mRNA expression ofPRMT1and DDAH1were meansured by RT-PCR.Results: The plasma concentrations of ADMA were significantlyelevated in the kidney of diabetic rat (314.70±40.85versus159.99±32.08μmol/L; P<0.01), and were significantly reduced by telmisartan (237.25±23.93versus314.70±40.85μmol/L; P<0.01).The mRNAexpression and the average optical density of PRMT1in DM group and TMgroup were much higher than NC group (P <0.01), however, the mRNAexpression of DDAH1was significantly decreased (P <0.01), After8weeks-treatment, the ADMA and PRMT1were decreased, while DDAH1and NO was increased, and the renal function was improved obviously.Conclusions: In diabetes condition, the expression of thePRMT1-ADMA-DDHA1axis in kidney tissue was abnormal. Telmisartanincreases the expression of DDAH1and decreases PRMT1in kidney, whichleads to the lower ADMA and reduces the oxidative stress and protects therenal function. |
