Objective:Used gene microarray to study differential gene expression profiles in invasive pituitary adenomas and noninvasive pituitary adenomas,to explore the potential invasiveness of pituitary adenomas associated genes at the genetic level.Methods:According to the judgment method of invasive pituitary adenomas, collect6tissue samples of invasive and non-invasive pituitary adenoma respectively in the neurosurgery of The First Affiliated Hospital of Chongqing Medical University, and divided each into3parts:one is delivered to the department of pathology of Chongqing Medical University for pathological examination; one is send to Shanghai Hualian biotechnology corporation for the Illumina gene expression in human whole genome microarray detect gene expression profiles in6cases of invasive pituitary adenomas and6cases of non-invasive pituitary adenomas, analysis of differentially expressed genes, and choose two differentially expressed genes validated by Reverse transcription polymerase chain reaction (reverse transcriptionpolymerase chain reaction, RT-PCR).Results:Gene microarray selected153differentially expressed genes, including63up-regulated genes and90down-regulated genes. Some of differentially expressed genes of cell adhesion, oncogenes, signal transduction related genes. On the up-regulated genes CDH12(N-cadherin2), and down-regulated genes KLF4(Kruppel-like factor4) line RT-PCR validation results was consistent with the microarray.CONCLUSION:Gene expression profiles of invasive pituitary adenomas are different from non-invasive pituitary adenomas, these differentially expressed genes to study the pathogenesis of invasive pituitary adenomas provide clues, which KLF4differentially expressed genes and SERPINE1the role of cell signal transduction may be through access to the invasiveness of pituitary adenomas play a certain role...
