As one of the most common benign tumor of the saddle area, pituitary adenoma is a tough problem clinicians have to deal with, in that it is difficult to whole resection, easy to recur, and frequent to lead to postoperational neurologic impairment and dyshormonism. Recently, gene therapy is becoming a promising treatment for pituitary adenoma in spite of reliable therapeutic targets deficiency. Therefore, it is significant to search the invasiveness relative regulatory gene of pituitary adenoma, and reveal the role it plays in the pathomechanism of occurrence and development of pituitary adenoma.Former researches uncovered that ebgabcer of zeste homolog 2(EZH2) overexpressed in many kinds of tumors such as liver cancer, breast cancer, endometrial cancer, prostatic cancer and gastric cancer, and had positive correlation to malignancy grade of the tumor. At present, EZH2 maybe play an important role in occurrence and development of pituitary adenoma, and become a new target of gene therapy. However, there is few research of EZH2 in pituitary adenoma.This research was divided into three parts to reveal the correlation between the expression of EZH2 and different levels of the invasiveness of pituitary adenoma(according to Knosp and Hardy-Wilson classification), and to discuss the role of EZH2 in occurrence and development of pituitary adenoma.Chapter 1: The expression level of EZH2 in pituitary adenoma.Objective: To research the expression of EZH2 in pituitary adenoma and the correlation between the expression of EZH2 and the invasiveness of pituitary adenoma.Methods: Immunohistochemistry was used to detect EZH2 expression level in 41 cases of pituitary adenoma from different ranks, then the expression level differences of EZH2 between different ranks of pituitary adenoma and the correlation between EZH2 and the invasiveness of pituitary adenoma were analyzed via rank sum test(SPSS).Results: There were no significant correlations between the expression level of EZH2 and the patients’ age and gender, but the EZH2 expression level had a significant correlation with the invasiveness of pituitary adenoma(P=0.002), and the higher invasiveness rank the pituitary adenoma belonged to, the higher expression level of EZH2 was detected.Conclusion: The expression of EZH2 has significant relevance to the invasiveness of pituitary adenoma and rise with the invasiveness rank, but not to the patients age and gender,.Chapter 2: The expression of EZH2 and VEGF and their mutual relation in pituitary adenomaObjective: To research the correlation between the expression level of VEGF gene and EZH2 in different ranks of pituitary adenoma.Methods: VEGF was detected in 41 cases of pituitary adenoma from different ranks via immunohistochemistry, and Spearman correlation coefficient method was applied to analyze the correlation between EZH2 and VEGF in pituitary adenoma.Results: The analyzation showed that the expression level of VEGF had significant correlation with the invasiveness of pituitary adenoma(p=0.006), Moreover, there was linear positive correlation between EZH2 and VEGF(r=0.531,p<0.001).Conclusion: The expression of VEGF has significant relevance to the invasiveness of pituitary adenoma, and a linear positive correlation exists between EZH2 and VEGF, both of which maybe affect the invasiveness of pituitary adenoma.Chapter 3: The expression of EZH2 and PCNA and their mutual relation in pituitary adenomaObjective: To research the correlation between the expression level of PCNA gene and EZH2 in different ranks of pituitary adenoma.Methods: PCNA was detected in 41 cases of pituitary adenoma from different ranks via immunohistochemistry, and Spearman correlation coefficient method was applied to analyze the correlation between EZH2 and PCNA in pituitary adenoma.Results: The analyzation showed that the expression level of PCNA had significant correlation with the invasiveness of pituitary adenoma(p=0.004), Moreover, there was linear positive correlation between EZH2 and PCNA(r=0.608,p<0.001).Conclusion: The expression of PCNA has significant relevance to the invasiveness of pituitary adenoma, and a linear positive correlation exists between EZH2 and VEGF, both of which maybe affect the invasiveness of pituitary adenoma.
