Objective: In our Preliminary studies, we first discovered EMS1over-expression in gastric cancer by gene chip. And we confirmed that EMS1couldenhance MGC803cells proliferation, invasion and migration. Purpose of this study isto detect silencing EMS1expression by siRNA on adhesion of MGC803, and toexplore the specific molecular mechanism.Method:1. Adoption of cell-cell adhesion and cell-matrix adhesion test detects theadhesion of MGC803cells that after silencing EMS1expression.2. Gene expression in MGC803and MGC-803-EMS1were detected by4×44KAgilent whole Genome Microarray. Filter out with fold change≥2as obviousup-regulated genes and fold change≤0.5as obvious down-regulated genes.3. Real-time PCR and Western Blotting were used to confirm the results of genechip.4. Analyze differences expression genes by genetic analysis software.Result:1. Cell-cell adhesion test showed that the adhesion of MGC-803-EMS1cellwas lower than MGC803and MGC-803-NEU(P<0.05).2.
