Objective: To investgate the association between the subclasses of high densitylipoprotein (HDL) and APOA5gene-1131T>C polymorphism inhypertriglyceridemic patients.Method: APOA5-1131T>C gene polymorphism was assayed in109hypertriglyceridemic patients (60.31±12.12years) and117healthy subjects (62.94±12.04years) by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP), the concentrations of apoA5, LCAT and CETP in plasma weremeasured by ELISA Kits, the subclasses of plasma HDL were determined bytwo-dimensional gel electrophoresis conjunction with immunodetection method,plasma lipids and apolipoproteins were detected by automated biochemical analyzer.Results:1. The levels of plasma TC, TG, LDL-C, apoB100, CETP, preβ1-HDL, HDL3bandHDL3awere significantly increased in hypertriglyceridemic patients comparedwith control, while HDL-C, apoA1, apoA5, HDL2b, HDL2aand LCAT weresignificantly decreased (P <0.01).2. The frequency of-1131TT genotype and T allelic in APOA5gene ofhypertriglyceridemic patients were lower than those of control group (47.7%vs50.4%,67.4%vs71.75%, P <0.05), while the frequency of-1131CC and C allelicwere higher than those of control group (12.9%vs6.9%,32.6%vs28.25%, P <0.05).3. Both in hypertriglyceridemic group and control group, apoA5levels in carriers ofTT and TC genotype were significantly decreased compared with TT carriers (P <0.001). The levels of apoA5in CC carriers in hypertriglyceridemia wereapparently lower than that of controls (P <0.001).4. The contents of preβ1-HDL and HDL3awere significantly increased in CC carriers compared with TT and TC carriers (P <0.01), but the levels of HDL2aand HDL2bwere significantly increased (P <0.01) in both hypertriglyceridemia and control.5. All subjects were divided into high (apoA5≥276.83mg/L), middle (153.59≤apoA5<276.83mg/L) and low (apoA5<153.59mg/L) concentration groupsaccoding to apoA5concentrations. Preβ1-HDL and HDL3awere significantlyelevated (P <0.01,P <0.05), while HDL2awas decreased significantly (P <0.01),with the decreasing of apoA5level.Conclusion: ApoA5concentrations are reduced in HTG, and APOA5-1131C allele isassociated with the decrease of apoA5, which might influence the changes of HDLsubclasses distribution.
