Effect Of "Treating The Lungs And The Intestine Simultaneously" On The Expression Of SPA And P38in Lung And Large Intestine Tissue Of ALI Rats

Objective:By observing the expression of pulmonary surfactant protein A(SPA) and P38after intragastric administration with qidonghuoxueyin (treating lung),dahuangtang(treating inteseine), qidonghuoxueyin mixed with dahuangtang (treating the lungs and the intestine simultaneously) respectively, this study evaluated the effects of different chinese traditional medicine treatment, and provide the basis for the "treating the lungs and the intestine simultaneously" theory.Method:72SD rats were randomly divided into six groups:normal saline group(NS group), model group (M group), qidonghuoxueyin group(Q group), dahuangtang group(D group), Dexamethasone group(DXM group) and qidonghuoxueyin mixed with dahuang group(QD group). The ALI rat modle was built by instillation with lipopolysaccharide directly into the airway while the NS group with NS. The intragastric administration was performed in each group with the Corresponding drug at the fixed time points:4hs,12hs before model-building and12hs after model-building. All the rats was killed24hs after model-building, and the necessary organs were harvested. The expression of SPA in bronchoalveolar lavage fluid(BALF), lung and large intestine was detected through enzyme-linked immunosorbent assay(ELISA). The expression of P38in lung and large intestine was detected though western blot.Result:1) The content of SPA in lung and BALF of NS group was higher than other groups(P<0.05) while in the M group was lower than other groups(P<0.05). The content of SPA in lung of QD group was higher than D and Q groups(P<0.05). The content of SPA in lung of DXM group was higher than Q and D groups(P<0.05), but has no significant difference with QD group.2) In large intestine, the content of SPA of M,D,Q,QD and DXM groups was rising gradually and the SPA of NS group was higher than Q,D and M groups (P<0.01). The SPA of M group was lower than NS,DXM and QD groups (P<0.01). The SPA in QD group was higher than Q,D groups,with no significant difference.3) In lung,the P38of M group was higher than NS,DXM,Q and QD groups (P<0.01),higher tnan D group (P<0.05).The P38of QD group was lower than D and Q groups,and has significant difference with D group (P<0.01).There was no significant difference between DXM and QD groups.4) In large intestine, P38of M group was higher than NS,DXM and QD groups (P<0.01),higher than D group (P<0.05). The P38of QD group was lower than D and Q groups, and has significant difference with Q group (P<0.05). There was no significanr difference in NS,QD and DXM groups.Conclusion:This study showed there is relevance between the extent of lung injury and the content of SPA in lung and BALF. The content of SPA was decreased and the expression of P38was increased in lung and large intestine during ALI. The Chinese traditional medicine treatment ameliorate the lung injury condition by increasing the content of SPA, and decreasing the expression of P38. The QD group showed better effect than Q and D groups in the lung tissue section, content of SPA, expression of P38, but has no significant difference with DXM group.
