Objective To investigate the expression of the PI3K/AKT protein downriver signalconduction circuit PI3K and the AKT activation form P-AKT and endoplasmicreticulum (ER) enzyme ENTPD5in different stages of treatment in acute leukemia (AL)and to explore PTEN gene expression in acute leukemia, the expression and itsrelationship with acute leukemia and the prognosis.Methods The cases included111patients with AL:78AML cases (48denovo cases,10relapse cases,20AML-CR cases),33ALL cases(14denovo cases,9relapse cases,10ALL-CR cases);62denovo patients with AL:26high-risk cases,19standard-riskcases,17low-risk cases. Western blot was employed to detect PI3K, and the AKTactivation form P-AKT and ENTPD5expression levels in different stages of treatmentin acute leukemia (AL), comparison in different stages of treatment and in low-risk,standard, high-risk group of acute leukemia amount of protein expression. Polymerasechain reaction (PCR) to detect the expression of PTEN gene in111cases of acuteleukemia (AL), also did analysis on the expression of PTEN relativity with clinicalparameters.Results PI3K, P-AKT and ENTPD5expression levels decreased significantly in ALinitial treatment than in AL-CR (P<0.05), Different risk stratification of patients, thereare the three protein expression differences, and the difference was statisticallysignificant (P<0.05). In111patients with AL,34cases of PTEN gene were not detected.In111diagnosed AL patients the complete remission rate (CR) of PTEN negative waslower than that of PTEN positive(P<0.05). The negative of PTEN had certain relativity with peripheral blood leukocyte count and the proportion of progenitor cells bonemarrow in pathogenesis.Conclusion The activation of PI3K/Akt pathway may play important roles in thedevelopment of AL, at the same time endoplasmic reticulum (ER) enzyme ENTPD5istherefore an integral part of the pathway and also play important roles in thedevelopment of AL. It seemed that expression of three proteins may be used as a markerof poor prognosis in acute leukemia.Gene PTEN is very important in the developmentof the acute leukemia and the pathogeny of acute leukemia. PTEN gene participate inthe development of acute leukemia.Detection of PTEN gene could be used as indicesfor judging prognosis....
