Objective:To investigate the expression of PTEN and integrin-linked kinase (ILK) in squamous cell carcinoma of the eyelid and conjunctiva and the degree of differentiation, as well as their inter-relationship. And to explore the clinical significance and effect in the development of squamous cell carcinoma of the eyelid and conjunctiva.Methods:We collected34cases of squamous cell carcinoma of the eyelid and conjunctiva, the corresponding adjacent and normal tissues, All the tissues were fixed in4%paraformaldehyde,conventional dehydration and embedded in paraffin. The expression of PTEN protein and ILK were detect by EliVisionTM plus immunohistochemical stain in34cases of squamous cell carcinoma of the eyelid and conjunctiva.Results:1、The expression results of PTEN protein and ILK in squamous cell carcinoma of the eyelid and conjunctiva and the corresponding adjacent and normal tissues PTEN protein in squamous cell carcinoma of the eyelid and conjunctiva and the corresponding adjacent and normal tissues were23.53%(8/34),100.00%(10/10), the expression of PTEN protein in cancer group was significantly lower than the adjacent and normal group,the difference was significant(P=0.000,P<0.01).ILK in squamous cell carcinoma of the eyelid and conjunctiva and the corresponding adjacent and normal tissues were73.53%(25/34),0.00%(0/10), the expression of ILK protein in cancer group was significantly higher than the adjacent and normal group,the difference was significant(P=0.000,P<0.01).2. The relationship between the PTEN protein and ILK with the degree of differentiation Expression of PTEN protein and ILK weren’t correlated with age,sex and was no significant difference (P>0.05)。23cases of well-moderate group, ILK expression in14patients with positive and expression of all positive in11cases of poorly group,the difference was significant(P=0.017,P<0.05).3、The relationship of PTEN and ILK34cases of squamous cell carcinoma of the eyelid and conjunctiva, PTEN expression in8patients with positive and negative in26cases. ILK expression in25cases of positive and negative in9cases。 Expression of PTEN protein and ILK positive in3cases, were tested negative in6cases; PTEN protein expression in26cases negative, ILK,19were positive, ILK expression in10cases negative, PTEN protein,7cases were positive. The Spearman rank correlation analysis, with a negative correlation between the expression of (r=-0.469, P=0.005,P<0.05)。Conclusions:1. PTEN was expressed high in squamous cell carcinoma of the eyelid and conjunctiva, but shows little expression in the corresponding adjacent and normal tissues of the eyelid and conjunctiva. There was a positive correlation between the expression of PTEN and the tumor pathological grade, which may play an important role in the occurrence and development of squamous cell carcinoma of the eyelid and conjunctiva.2. ILK was expressed high in squamous cell carcinoma of the eyelid and conjunctiva, but shows little expression in the corresponding adjacent and normal tissues of the eyelid and conjunctiva. There was a positive correlation between the expression of ILK and the tumor pathological grade,which could be marker of tumor and to evaluate prognosis in squamous cell carcinoma of the eyelid and conjunctiva.3. The expression of ILK was negative relation with PTEN, and they can cooperate with in the occurrence and development of squamous cell carcinoma of the eyelid and conjunctiva. Up-regulated expression of ILK and down-regulated expression of PTEN may play an important role in the process of carcinogenesis and differentiation of squamous cell carcinoma of the eyelid and conjunctiva; ILK may be useful targets for treatment of squamous cell carcinoma of the eyelid and conjunctiva.
