Objective Filter out effective monitoring of peripheral blood of liver cancer cellssensitive indicators, degree of proliferation of hepatoma cells in the peripheral and theportal vein in patients with hepatocellular carcinoma in the assessment of the twoproceduresMethods Use fluorescence quantitative PCR (FQ-PCR) to detect the expression levelsof AFP mRNA，h-TERT mRNA and VEGF mRNA in40cases with HCC，20cases withliver disease and10cases with normal medical groups in the peripheral blood.58casesHCC patients underwent liver neoplasm resection randomly by advance regionalblocked blood flow(experimental group32cases) and Pringle ’s blockingmethod(control group26cases),and draw all patients’ peripheral blood(preoperative30min,postoperative30min，postoperative1d) and portal vein blood(before operationand after tumor resection),use Real-time FQ-PCR to quantificationally test AFP mRNAand h-TERT mRNA in sample.Results Three indicators of expression in HCC patients were significantly higher，thedifference was statistically significant (P <0.01)。Three indicators in HCC patients withpositive rates were77.5%，85%and72.5%，compared with the control group thedifference were statistically significant (P <0.01)。And the AFP mRNA，VEGF mRNAexpression in HCC patients with positive serum AFP levels related (P>0.05)，h-TERTmRNA and serum AFP levels there is correlation (P=0.01); three indicators for HCCpatients with TNM sub-period highly correlated (P <0.01)。The specificity of AFP mRNA in the diagnosis of HCC was higher，while the AFP mRNA and h-TERT mRNAjoint detection better.And over the same period,compare experimental group withcontrol group，AFP mRNA,h-TERT mRNA in peripheral blood has no obvious differentin preoperative,but the gene expression displayed a marked difference inpostoperative(P＜0.05)，The two groups of postoperative30min data respectively were241.65±46.52、848.96±160.08；797.27±165.36、2851.59±595.92. Before and after tumorresection the Index expression of portal blood respectively were：2214.47±334.16、8707.63±1318.60；578.03±83.48、2192.64±327.36. Control group respectively were：3454.19±598.25、13499.70±2403.56；1947.11±278.97；609.42±1062.41，it has astatistically significant differences(P＜0.05).Conclusion1. Join the detection of AFP mRNA and h-TERT mRNA in HCC diagnosis can increasethe sensitivity and specificity.2. AFP mRNA and h-TERT mRNA of peripheral blood and portal vein blood has apositive correlation in intraoperative surgical operation of liver cancer,this may be oneof the reasons how tumor cells transfer.3. It may have a blocking function for the spread of tumor cells by the advance regionalblocked blood flow, and more conducive to short-term recovery of patients.
