Objective To explore the relationship of the pathogenesis of rheumatic heart diseaseand MAPK signaling transduction pathway through the comparison and investigation ofthe expression of MAPK signal transduction pathway resection surgery of rheumaticheart disease and non-rheumatic heart valve disease.Methods1.General information Selected50cases of rheumatic heart valve patientshospitalized in July2010to November2011in the First Affiliated Hospital of AnhuiMedical University, Cardiac Surgery II Ward for surgery and complete information asthe experimental group;16cases of non-rheumatoid valvular patients as the controlgroup,included: ischemic heart disease with valve disease(chordal rupture andprolapse),pure vascular lesions involved the valve disease (aortic aneurysm or aorticdilatation),congenital heart disease (CHD) patients combined with valvular disease.Collected the valves of two groups of patients undergoing valve specimens and savedthe spare.2. Detection methods:2.1Used the method of Real-time polymerase chain reaction (real time polymerasechain reaction, RT-PCR) to quantitative analysis messenger ribonucleic acid (messageribonucleic acid mRNA) of three signal transduction pathways including P38MAPK,JNK, ERK in the both groups diseased heart valve tissue;2.2Used the method of Western blot to quantitative analysis the total protein andphosphorylation protein of three signal transduction pathways including P38MAPK,JNK, ERKin the both groups diseased heart valve tissue. 3. Statistical analysis: SPSS13.0statistical package for data analysis, statisticalmeasurement data value is expressed as: mean±standard deviation(x±s), comparingthe significant difference of both groups used t test.Results1RT-PCR: MAPK signal transduction pathways including P38MAPK, JNKand ERK mRNA expression in the two sets of valve is not present statisticaldifference(P>0.05);2Western-Blot: Total protein expression of P38MAPK, JNK and ERK three signaltransduction pathways in two sets of valves were no significant difference(P>0.05).Thephosphorylation protein expression of JNK signal transduction pathway wassignificantly higher in the experimental group valves than in the control group,and itwas significant difference (P <0.05), but P38MAPK ERK two signal transductionpathway phosphorylated proteins expression in two sets of valves have not existed asignificant difference (P>0.05).Conclusion MAPK signal transduction pathway has played a certain role in thepathogenesis of heart valve disease.The JNK signaling pathway of the MAPK signaltransduction pathway has played a important role in the pathogenesis of rheumatic heartvalve disease and P38MAPK, ERK two signaling pathway played a important role inthe pathogenesis of rheumatic heart disease have not yet conclusive. In this study, forin-depth exploration of the expression of the inflammatory signal transduction pathwaysin the pathogenesis of rheumatic heart disease, provide a potential intervention target forfuture prevention and treatment of rheumatic heart valve disease.
