Objective Neuron-specific-enolase (NSE) and soluble-protein100(S-100) immunhistochemical staining was performed in removal pavilion and neck intestinal segments of Hirschsprung’s disease (HD) and Allied Hirsschsprung’s disorder (HAD) to observe the expression characteristics of ganglion cell and nerve fibers, and to probe the differential diagnosis and pathomorphology characteristics of HD and HAD.Method Retrospective study combined with clinical data, to investigate removal pavilion and neck intestinal segments from45samples with HD, including37male and8female, ranging from4months to5years in our hospital during2005to2011, they are patients who have undergoing radical operation were stained by HE conventional method. The morphological and quantity change of the neuron and ganglion cell of the lesion were observed compared with the normal control group. There are24samples be determined the HD,9of them have complication in follow-up. They are defined group A. The left21samples is suspected group, among them there are16different kinds of complications. We define it group B. Select5normal removal colon wall edge of adult colon cancer as normal control group, we regard it as group C, NSE and S-100protein immunohistochemical markers were stained among these3groups’ganglion cell to observe the expression characteristics of neck and pavilion of group A and B and colon wall of group C, and analyze it combine with follow-up condition.Results (1)For group A, we can’t see any positive ganglion cells in NSE of neck segment colon wall, the majority of nerve fibers express positive, few of them weakly positive, S-100in neck segments of intestinal express strong positive reaction, nerve fibers proliferated markedly and disturbance in distribution, no "cell-form blank zone", here.(2) For group B, we can see NSE ganglion cell express strong positive reaction in neck segment colon wall. We can see a few of "cell-form blank zone" when S-100was stained.(3)For group C, the NSE immunohistochemical positive staining neuroganglion cells with heterogeneous color could be found in the neuroplexus of the colon wall in the control group with neurofiber distributed uniformly in the different layers of the colon wall, paralleling with the muscle fiber. We can see normal distribution of "cell-form blank zone" when S-100was stained.(4) The expression differences of NSE and S-100in neck segments of intestinal between group A and B have statistics meaning and there is some difference compare with the normal control group C.Conclusion1、NSE and S-100are markers with high ganglion cell specificity, which can be used as an important measure to judge the development of enteric nervous.2、The HD neck segments of NSE, S-100, to determine the expression of the ganglion cells and nerve fibers to grasp the pathophysiologic characteristics of HD, the further differential diagnosis of HD and HAD are helpful of choosing clinical surgery, in order to reduce the postoperativebloating, constipation and other complications, To improve the prognosis of the patients HAD children...
