Objective:Make a study of the active ingredients that have anti-tumor effect in Venenum Bufonis, screened out of the most appropriate proportion of the three toad poison drugs.Methods:The method of MTT and writhing with acetic acid were used to determine the cell growth inhibition on BEL-7402hepatoma cell lines and the analgesic effect of the mice of Bufalin, cinibufagin and resibufogenin at different concentration and different time. and to explore the role of resibufogenin to Bufalin, cinibufagin. Ultimately, Using orthogonal test method Studied compatibility of the three toads poison drugs about the anti-tumor effect and analgesic effects.Results:①The IC50values of BF, CBG、RBG in the role of cells after24h are:1.560×10-8、1.393×10-6、8.128×10-5, B5、CBG、RBG can significantly inhibit the proliferation of BEL-7402human hepatoma cells, three toads poison drugs have a clear dose and time effect relationship on hepatoma cells, and inhibitory effect increased With the extension of time or the increase of concentration. In the mouse writhing analgesic experiments, the inhibition of them significantly increase with the increase of concentration.②combination of two of the different ratio of inhibition has little effect on BEL-7402hepatoma cell lines,there is no significant differences with separate inhibition. compatibility between the components on the analgesic show great effect. duing to the proportion of different the compatibility of the analgesic effect show the impact of suppression and the promotion.③The orthogonal experiment results show that the inhibition of the three components in lower concentrations in a1:1:1ratio compatibility is better on BEL-7402human hepatoma cells; analgesic effect of mice, when RBG is at higher concentrations, the ratio of RBG、CBG and BF is4:2:1(0.52:0.219:0.11), the effect is better.Conclusions:The inhibition on BEL-7402human hepatoma cells and analgesic effects of three toad poison drugs showed that BF has the best effect, followed by the CBG, the RBG is weakest. This study screened a more appropriate ratio of compatibility on anti-tumor and analgesic effect, and at the same time found that the effect of compatibility and separate the effect were of less difference. Therefore, in the case of higher monomer separation technology requirements, We can combine the actual industrial production or new drug R&D needs, chose of the ratio of compatibility of active ingredient groups in Venenum Bufonis.
