OBJECTIVE: In order to improve the formulation of traditional Chinese medicinehuchangqingdu soup and improve their targeting, we preparated the colon targeted huchangqidupellets,and determined the process conditions，investigated the stability of formulations. So that itcan achieve purposes of targeted drug delivery,and attenuated efficiency.METHODS: Based on the study of single factor, we choose the orthogonal test to optimize theextraction process Chinese medicine,in order to provide high-quality raw materials for furtherpellets preparation; We choose extrusion–spheronization to prepare Huchangqingdu pellets bythe conbined methods of single factor text and orthogonaltest, in order to realize the whole degreeand yield of the comprehensive evaluation index to optimize the prescription and preparationprocess; examine its micromeritic nature; coating and excipients to achieve colon targeting of thepellets.initial inspection visits and targeted Huchangqingdu pellets preparation in vitro dissolutionand in rats in vivo pharmacokinetics using HPLC.RESULTS: The optimum extraction process is the3times of extraction,8times of addingwater, and extraction time is2hours. The best prescription for Pellets is that the proportion ofdrug, pectin calcium and microcrystalline cellulose is1:1:2.
