Background and objective:colorectal cancer is one of the most common malignancies. It is merely next to lung cancer and reside the Second with high morbidity and mortality. Its incidence and mortality rates were43.35/100000and22.42/100000. Since recent methods of treatment are not ideal, therefore, early detection, early diagnosis, early treatment and the development of standardized operation treatment and adjuvant chemoradiotherapy remains to be the key of improveing therapeutic efficacy of colorectal cancer. With the emergence of new personalized cancer treatment and the occurrence of gene targeted therapy concept, Studying the expression pattern variation of the colorectal cancer related genes in the molecular level may provide new ideas for effective early colorectal cancer diagnosis and treatment, and may also contribute to explore the prognostic factors of colorectal cancer, thus improving the treatment effect of colorectal cancer. HPRG1is a recently discovered gene which contains a Lish/CTLH domain. Lish domains are found in the product of many cancer related genes. It has been reported that HPRGl regulated cell physiology via MAPK signaling pathway and TGF-β signaling pathway which play an important role in the occurrence and development of cancer cells. This paper aims to study the expression variation and the clinical significance of HPRG1in colorectal cancer, and provide new target points of research for the early diagnosis and treatments of colorectal cancer.Materials and methods:60colorectal cancer tissue samples without preoperative radiotherapy, chemotherapy were collected from gastrointestinal surgery in Xiangya No.2Hospital from June to December2011. In each case, adjacent normal tissue of colorectal cancer tissue was set as control group. RT-PCR was used for detection of the expression difference of HPRG1in colorectal cancer tissue and its adjacent normal tissue in m-RNA level. Moreover, the expression difference of HPRGl in protein level was detected by Western blot, and immunohistochemistry was used for further detection of the expression difference of HPRG1in colorectal cancer tissues and its adjacent normal tissue in tissue level.Results:The expression of HPRG1in colorectal cancer tissues is significantly higher than that of adjacent normal mucosa tissues whose distance from the cancer tissue are greater than5cm (P<0.05). The positive expression rate is related with the cancer cell differentiationdepth of invasion, lymphatic metastasis and distant organ metastasis (P<0.05), while is not relevant to patients’ gender and age (P>0.05).Conclusion:The high expression rate of HPRG1in colorectal cancer is related with the cancer cell differentiation,depth of invasion, lymphatic metastasis and distant organ metastasis, while is not relevant to patients’ gender and age.
