Objective:To investigate NF-κB interacting protein in human myometrium smooth muscle in no-labor and labor situation.Methods:the expression of NF-κB was detected by Western blotting: co-immunoprecipitation and sodium dodecyl sulfate polyacrylamide gel electrophoresis was performed for the separation and purification of proteins interacting with NF-κB in labor and no-labor situation; liquid chromatography-tandem electrospray mass spectrometry and profile analysis was used to identify the separated specific protein and their possible function.Results:positive expression of NF-κB was detected in the smooth muscle of lower uterine segment from both natural term delivery patients and no-term delivery patients.12specific proteins were identified by liquid chromatography-tandem electrospray mass spectrometry and profile analysis, including cell structure associated protein, calcium binding protein, Acute phase reactive protein and signal transduction associated proteinConlusion:Utilize co-immunoprecipitation and mass spectromelry, Twelve kinds of proteins interacting with NF-κB are identified in the smooth muscle of lower uterus segment from term natural labor patients and no-term labor patients. They are Desmin. EH domain-containing protein2.Heat shock70kDa protein.Annexin A6.Alpha-actinin-1. Filamin-A,TRIM27Isoform Beta of Zinc finger protein RFP, Vimentin, ATP synthase subunit alpha,mitochondrial,Isoform1of Alpha-1-antitry-psin, Tubulin alpha-lA chain, Tubulin alpha-1C chain, ning kings of proteinsthree kinds of proteins interacting with NF-κB are identified in term labor patients and no-term labor patients.but term labor patients have more than three kinds of proteins:Alpha-actinin-1. Filamin-A、 TRIM27Isoform Beta of Zinc finger protein RFP.It suggest these proteins play a role in parturition initiation involving NF-κB.
