A Study About Proteins Interacting With NF-κB In The Smooth Muscle Of Lower Uterine Segment From Term Natural Labor Patients And Drug Induced Labor Patients

Objective:In the smooth muscle of lower uterine segment from term natural labor patients and term drug induced labor patients, to identify the proteins interacting with NF-κBMethods:1. Collecting smooth muscle of lower uterine segment from term natural labor patients and term drug induced labor patients, to identify whether NF-κB p65protein in the smooth muscle by Western blotting.2. Then, co-immunoprecipitation with antibody NF-κB p65protein, sodium dodecyl sulfate polyacrylamide gel electrophoresis was performed for the separation and purification of complexes containing NF-κB p65protein.3. Analysis and identify the protein interacting with NF-κB by liquid chromatography-tandem electrospray mass spectrometry and database.Results:1. NF-κB p65protein was detected in the smooth muscle of lower uterine segment from both natural term delivery patients and drug induced term delivery patients.2. Using sodium dodecyl sulfate polyacrylamide gel electrophoresis to Separated co-immunoprecipitation complexes. Protein band with molecular weight of72KD was detected in the experimental group, but not in the control group.3. Seven specific proteins were identified by liquid chromatography-tandem electrospray mass spectrometry and profile analysis. According to their function, these protein can be devided into3group:cell structure proteins, heat shock proteins and signal transduction proteins.Conlusion:Seven kinds of proteins interacting with NF-κB are identified in the smooth muscle of lower uterus segment from term natural labor patients by Peptide mass fingerprinting and Co-Immunoprecipitation. They are desmin, filamin-A, actinine, zinc finger protein, Heat shock protein70, Annexin A6, EH-domain protein2. Meanwhile, five kinds of proteins interacting with NF-κB are identified term drug-induced labor patients. They are desmin, filamin-A, actinine, Heat shock protein70, Annexin A6. The two groups share the same five kinds of proteins, there are two kinds are different.It suggest these proteins play a role in parturition initiation involving NF-κB.
