Ganoderma lucidum (Fr.) Krast (Polyporaceae), one of the most famous traditional Chinese medicinal mushrooms, is a popular folk and oriental medicine used to treat many diseases.In this research, Ganoderma lucidum was extracted by70%MeOH, and then was suspended in water and successively partitioned and extracted with petroleum ether, ethyl acetate and n-butanol. The three extracts were separated by different chromatographic methods (such as Silica gel column, RP-18, Sephadex LH-20, atc). Thirteen compounds were identified as Ergosterol-7,22-diene-3β-ol (1), Ergosterol peroxide:5α,8α-epidioxyergosta-6,22-dien-3β-ol (2), Ganodermanondiol (3), Ganodermadiol (4), Cerevisterol (5), Methyl ganoderate D (6), Lucidumol B (7), Ganoderiol F (8), Ganoderiol A (9), Ergosta-7,22-diene-3β,5a,6β,9α-tetraol (10), Ganodermanontriol (11), Methyl ganoderate B (12), Lucidenic acid LM1(13).To explore whether the isolated compounds induced the expression of HO-1, we exposed Hep G2cells to varying concentrations of compounds1-13. The results showed that compound3inducing the highest amount of HO-1expression in a concentration-dependent manner.In this study, Ganoderma lucidum was extracted and isolated with seperation of chinese medicine looking for active compound. The study will lay the foundation for research and application of Ganoderma lucidum.
