Study On The Extraction Process Of Bioactive Constituents And Antitumor Chemical Compounds In Ganoderma Lucidum

Polysaccharide and ergosterol in Ganoderma lucidum were investigated in the thesis. The determination methods of them are set up and the optimal extraction process is also established. The results are useful for the further product development from Ganoderma lucidum.The determination method of polysaccaride: the determination of polysaccharide is established with method of phenol-vitriol, in which dextrose is the standard sample.The concentration is determined by spectroscopy at 485nm.The regression equations of the standard curves based on the concentrations(C) versus the absorptive value(A) are: A=10.559C+0.0019 (R=0.9991). The liner range is from 20μg to 120μg.The determination method of ergosterol: the contents of ergosterol in extractive solvent were determined by HPLC. The conditions of chromatogram follows as below: the column [kromasil - ODS C18(4.6×150mm;5μm)], the mobile phase (methanol), the speed (1ml/min), the temperature of column (25℃), it is detected at 282nm. The regression equations of the standard curves based on the concentrations(x) versus the square of the peak(y) are: y=32767x-233.91 (R=0.9996). The liner range is from 20μg to 400μg.The optimal extract process established through orthogonal design test for polysaccaride is to extract four times at 90℃with the ratio of 1:60 (m/v) for 3.5h.and the suitable one for ergosterol is to extract three times using alcohol(95%) with the ratio of 1:10 (m/v) for 1.5h..Twenty-two compounds were isolated from the body of Ganoderma lucidum by using column chromatography, including HPLC. Five of them were identified on the basis of spectroscopic studies. The five coumpounds are ergosterol(1), ganoderiol B(2), ergosterol peroxide (3), and the other two possible structures.In a screen for antitumor activity substances, the cytotoxic activities against human tumour cell were tested. Ergosterol, ganoderiol B and ergosterol peroxide showed cytotoxic activities.
