Objective: The aim of the study is to analysis therelationship between the expression of Cox-2,5-Lox and Bcl-2and theirhistopathological characteristics in gastrointestinal stromal tumors(gastrointestinal stromal tumors, GISTs). Then, to evaluate their clinicalsignificance. Method:100cases of paraffin block filed included in myresearch were selected from Department of Pathology, Affiliated Hospitalof Luzhou Medical College from Jan2009to Dec2011which weredevided into low risk group (28cases), moderate risk group(40cases)and high risk group (32cases). At the same time,20cases of normalgastrointestinal organization were taken out as control group. Histologicaltype of the samples was confirmed by pathological diagnosis. Afterreading the samples again, we detected the expression level of Cox-2,5-Lox, Bcl-2in GISTs and normal gastrointestinal tissue byimmunohistochemical SP method. Then we analysis the relationship ofthe three and the correlation between their expression and the clinicalpathological features in GISTs. The datas were analyzed by chi-squaretest, Fisher exact probabilities test, preason rank correlation analysisperformed by SPSS17.0. Results:(1) The positive rates of theCox-2,5-Lox and Bcl-2expressed in GISTs were68%,77%,85%,respectively, which were higher than normal organization group(15%, 20%,15%). The difference was statistically significant (p<0.05).(2) Thepositive expression rate of Cox-2,5-Lox, Bcl-2in GISTs wassignificantly increased in the group with high histologicalgrade(X~2=8.396,7.498,6.162, p<0.05). The difference between low andmoderate risk groups of Cox-2, low and high risk groups of Cox-2, lowand high risk groups of5-Lox, moderate and high risk groups of5-Loxwas significant(X~2=5.783,6.461,6.104,6.402, p<0.05). The result alsoreflected in low and high risk groups of Bcl-2(X~2=6.687, p<0.05).(3) Thepositive expression rate of Cox-2in GISTs had no significant correlationwith age, sexuality, tumor position,tumor size and cell nucleus mitosis(p>0.05). The positive expression rate of5-Lox in GISTs had nosignificant relationship with age, sexuality, tumor position,tumor sizeand cell nucleus mitosis (p>0.05). The positive expression rate of Bcl-2in GISTs concerned with the numer of cell nucleus (X~2=4.286, p<0.05),had no significant relationship with other clinical pathological factors(p>0.05).(4) The result of correlation of Cox-2,5-Lox and Bcl-2showedthat, Cox-2and Bcl-2in GISTs tissues had positive correlation (r=0.227,p<0.05).5-Lox and Bcl-2in GISTs tissues had positive correlation(r=0.275, p<0.05). The expression of COX-2had no significantrelationship with5-LOX(r=0.157, p>0.05). Conclusion:(1) Theexpression of Cox-2,5-Lox and Bcl-2in GISTs was significantly higherthan normal organization group, so the expression of the three may beplay an important role in the process of tumor occurrence and development.(2) The expression of Cox-2,5-Lox and Bcl-2uniteddetected in GISTs is a objective indicator of early diagnosis, distinguishinvasion and prognosis. Inhibition of Cox-2,5-Lox and Bcl-2specificallycan delay the development of GISTs and improve the prognosis.(3) Thepositive expression of Cox-2,5-Lox was correlated respectively withBcl-2. Cox-2,5-Lox may play an important role by the approach ofBcl-2. Cox-2had no significant relationship with5-Lox. It is need furtherresearches to prove that whether Cox-2and5-Lox have differentinfluence factors or uncorrelated relationship in GISTs.
