The Preliminary Study Of Anti-chronic Nephritis From Rostellularia Procumbens (L.)

Herba Justiciae is the whole dried plant of Rostellularia Procumbens(Lin.)Nees,also named as Liujiaoxian, Xiaoqingcao, Chiyan, Haiercao, Dayacao, Wuleicao, Maozelan, Jiejiehan and etc.China is mainly distributed in southwestern and Taiwan, Abroad,mainly distributed in Afri-ca.It has been used to qingrejiedu,dampness swelling and huoxuezhitong,It commonly used to treat cold and fever,cough,sore throat,Malnutrition,Shire xieli,malaria,jaundice,edema,urine pouringmuddy,musculoskeletal pain,br-uises,ulcer boils and eczema in clinical. Included in the Chinese Pharmacopoeia (1977edition, a). According to reports, it has function in treating chronic nephritis,but its material basis for the treatment of nephritis and its mechanism is not very clear,and should be in-depth research.In this thesis, using the system solvent,its ethanol extract is divided into petroleum ether, ethyl acetate, n-Butanol and aqueous solution of four fraction. Doing toxicological and pharmacological screening,obtain active fractions based on mesangial cells HBZY-1by MTT approach with LPS as stimulus.The results showed that petroleum ether, ethyl acetate and aqueous solution of three parts have the biological active for anti-nephritis.their IC50were187μg.mL-1,189μg.mL-1and720μg.mL-1。Adoptting silica gel column chromatography, gel permeation chromatography, MCI and HPLC column chromatography separation,19compounds were isolated from the activity franctions of petroleum ether extracts(590g).And17compounds were furtnerly identified as neojusticin A,neojusticinB,neojusticinC,TaiwanE,6’-hydroxy-justicidinB,β-sitosterol, cholesterol,(3-daucosterol and etc.Then Seven of these compounds was first isolated from this plant.They were pentadecanoic acid^cholesterol and five homolog(compound13-17).Similariy,doing toxicological and pharmacological screening,obtain active active monomers based on mesangial cells HBZY-1by MTT approach with LPS as stimulus.The results showed that compound17’s IC50were16.1μg.ml-1, compound15’s IC50were96.7μg.mL-1.It provide a scientific basis to clarify the material basis of anti nephritis. A more systematic quality analysis were laid a foundation for the evaluation of resource quality and the development and utilization.
