Study On The Chemical Constituents And Quality Analysis Of EtOAc Extraction From Rostellularia Procumhens (L.) Ness

Spreading over regions of southwestern of China and Taiwan withabundant resources, Justicia, which belongs to the Family Acanthaceae, is thewhole dried herba of Rostellularia Procumbens (L.) Nees. Being bitter in tasteand cold-natured, Justicia remains mainly used for the treatment of malaria,dysentery, jaundice, nephritis edema, bones and muscles sprain swellingdisease with effects like clearing heat and detoxification, diuretic swelling,malaria and others. As a traditional Chinese herbal medicine, Justicia hasbroad application prospects and it has been recorded in “the1977edition ofChinese Pharmacopoeia”. According to relevant reports, this medicine stillseparately needs to be further studied on its significant anti-tumor effects, forthe substance basis and mechanism of action haven’t been made clear.This project carried out a preliminary study of the active ingredient of theanti-chronic nephritis after getting the petroleum ether and ethyl acetate sitesby means of solvent extraction, finding that the two sites actively have certaininhibition against HBZY-1mesangial cells proliferation. Study of acetateextract on its contained chemical composition provided important theoreticalguidance and experimental basis for further development and clinicalapplication of Jusicia.The acetate extraction (195g) separated and purified by repeated Sephadex column chromatography, crystallization and recrystallizationmethod,17compounds were obtained, with the help of their physicochemicalproperties, NMR spectrum data and related literature,15of them wereidentified as follows: Neojusticin B（JC-1）、Neojusticin A（JC-2）、6′-hydroxyjusticidin B（JC-3）、Justicidin C（JC-4）、Chinensinaphtho（lJC-5）、6′-hydroxyjusticidin C（JC-6）、Taiwanin C（JC-7）、Neojusticin C（JC-8）、4’-demethyl-chinensinaphthol methyl ether（JC-9）、Palmitic acid（JC-10）、Stigmasterol（JC-11）、 Quercetin-7-O-α-L-rhamnopyranoside （JC-12）、 β-sitosterol（JC-13）、β-daucostero（lJC-14）, among which compound11and12were forthe first time isolated from Justicia.By using RP-HPLC chromatograph to conduct three lignan compounds asan index, this project contributed to the establishment of quality evaluationsystem of Justicia.
