The Inhibition Of Synovial Proliferation With Dang Gui Nian Tong Tang-plus And Methotrexate In AA Rats

Objective: To investigate the role of the Angelica Niantong Decoction andMethotrexate in rats with adjuvant arthritis (AA) synovial hyperplasia, observed HIF-1αwas the exrpession of VEGF and pathological rat synovial change. Methods:48wistarrats were randomly divided into normal group, model group, treatment group and controlgroup. The AA model was established and observed the dynamic changed arthritis throughthe activity indicators. After18days of the drug intervention, the ankle joint was takenand observed HIF-1α and VEGF with immunohistochemical staining and HE staining. Itwas analysised that the relationship between the expression of HIF-1α and VEGF and thearthritis activity indicators, synovial pathology in the synovial tissue. Results: Before thetherapy, the baseline was compared(P>0.05) that arthritis index, ankle swelling, joint painindex line. After the intervention, compared with the normal group, the treatment groupand control group could inhibit AA rat ankle joint swelling (P<0.05), to reduce the anklejoint pain index (P<0.05), HE stain reduce the synovial proliferation and inflammatoryresponses (P<0.05), the expression of HIF-1α and VEGF decreased (P<0.05) in synovialmembrane, and each index was positively correlated. Compared with the control group,the expression of factors in the treatment group was differences (P<0.05). Conclusion: Itwas show that Angelica Niantong Decoction and Methotrexate can inhibit HIF-1α andVEGF in the pathology score, joint swelling, joint pain index expression. Thus, it wasassumed that the ointment and methotrexate may regulate a series of downstream targetgenes VEGF by HIF-1α, this may be a way of inhibition of synovial hyperplasia.
