| ObjectiveTo copy the hormone refractory prostate cancer lung metastasis model to evaluate Cryoablation of surviving prostate cancer tumor angiogenesis, tumor invasion, cell proliferation and cell aptosis in prostate cancer, and the effect of the therapy with Recombinant Human Endostatin (Endostar).Materials and Methods120male BALB/c(nu-/nu-) mice with PC-3prostate cancer were established. Experimental mice were randomized into three groups:the AHCS therapy group (n=40), the combined therapy group(n=40), and the tumor-bearing control group (n=40). After the AHCS therapy. Residual cancer tissues of10mice in every group were obtained at cryotherapy,7d,14d and21d. VEGFã€MVDã€MMP-9〠Ki-67and Caspase3protein expression were detected by immunohistochemical staining at different time after the freezing.Results1VEGFã€MVDã€MMP-9Ki-67and Caspase3were increased in AHCS therapy group during7days and21days after AHCS(P<0.05).2VEGFã€MVDã€MMP-9were decreased in combined therapy group as time goes on, and as the sametime Ki-67was up at first and then down, and caspase3was not predominant changed(P<0.05).3AHCS therapy group was in comparison with control group, There were significant change in VEGFã€MMP-9ã€Caspase3protein expressions at different time (P<0.05), and MVD and Ki-67were significant changed after14days on AHCS(P<0.05).4Combined therapy group was in comparison with AHCS therapy group, MMP-9was dominantly down at different time point(P<0.05). VEGFã€MVD went down after14days on AHCS(P<0.05).Ki-67went down from21day(P<0.05).Caspase3were increased in comparison with AHCS therapy group, but not statistic difference(P>0.05). ConclusionRemnants of the AHCS for prostate cancer, angiopoiesis, tumor invasion and cell proliferation gradually incresed over time; Cryoablation and combined Endostar decreased in angiopoiesis, invasion and cell proliferation, and increased in apotosis of tumor cells over time; Combined Cryoablation with Endostar are complementing each other, and the longer the duration of the application, the more obvious. |
