Object:1.To establish the model of EAT by Tg and C57BL/6mice.2.Isolated and cultured BMSC from Balb/c mouse compact bone through our published method.3.Based on the model of EAT,study the therapy of allogeneic bone mesenchymal stem cells(ABMSC) on the model and the relationship between the therapy and CD4+CD25+Treg cells.Materials and Methods:1.Isolated and cultured MSC from healthy Balb/c mouse compact bone through our published method. The immunophenotypic features of the cells were analyzed by flow cytometry.2.we selected36C57BL/6female mice about5weeks old,then they were divided into3groups..The A group was BMSC treatment group,The B group was model control group,The C group was normal control group.On0and14day, respectively every mouse in A group and B group was immunized by injection of150μg porcine Tg (pTg),every mouse in C group was by injection of saline water into the muscle in the two back vola pedis.3.On0,7,14and day every mouse in A group received3×105BMSC by tail intravenous injection.4.On28day,took blood,then mice were killed,put the thyroids in10%formalin immediately did flow cytometry and determined the TT3, TT4, TPOAb, TmAb, TgAb by optical light method.Results:1. TT4of A group and B group was significantly lower than C group(P<0.05),TPOAb,TmAb,TgAb of A group and B group significantly was higher than C group(P<0.05),TT3had no statistical difference in3groups(P>0.05).2.After the treatment,TPOAb, TmAb, TgAb of B group was significantly higher than A group(P<0.05),TT4of A group was higher than B group(P<0.05).3. The frequency of CD4+CD25+Treg cells of A group and B group was significantly lower than C group(P<0.05),After the treatment,B group was below the A group (P<0.05). 4. The frequency of CD25+CD4+Treg cells was negatively correlated with TPOAb, TmAb, TgAb in every group.5. A microscope to C group, thyroid follicular was mature, it was full of colloid, uniform distribution,the arrangement of follicles was lobular, The inflammatory cell was not seen in group,but seen little inShrinking follicles could be seen little in A group.thyroid gland flocculus of B group,lymphocytes invaded thyroid gland,9cases.Conclusions:1.TPOAb, TmAb, TgAb of model control group significantly higher than normal control group,lymphocytes invaded thyroid gland,those suggested that mice EAT was of success.2.TPOAb, TmAb, TgAb of B group significantly higher than A group,A microscope to A group the inflammatory cell was little seen. Shrinking follicles could be seen little in thyroid gland flocculus of B group, lymphocytes invaded thyroid gland,they above suggested that using MSC to mice EAT was effective.3.The frequency of CD4+CD25+Treg cells of A group and B group was lower than C group,The frequency of A group was below the C group,.we could conclude that the effects of BMSC on mice EAT may be by regulating the number of CD25+CD4+Treg cells.
