Object:1. Make use of C57BL/6mice to establish the model of hashimoto’s thyroiditis (HT) of mice, which is also named experimental autoimmune thyroiditis (EAT), has characters of increased antithyroid autoantibodies lymphocytes infiltrates thyroid.2. Separate and culture healthy C57BL/6mice’s (bone marrow mesenchymal stem cells)BMSCs, EAT mice received tail vein injection of BMSCs and compare the serum levels ofTT3、TT4、TgAb、TPOAb、TMAb and lymphocytes infiltration of the thyroid between models and control group, explore the immune modulator function of homologous BMSCs.3. Compare the serum levels of IFN-γ、IL-17between models and control group, analysis the correlation between IFN-γ、IL-17and TPOAb, in order to Explore the pathogenesis of HT and the immune regulation function of BMSCsMaterials and Methods:36female C57BL/6mice,all were6weeks old, the body weight were (20±1) g, divided into3groups randomly, model and treatment group (group A), simple model group (group B), and normal control group (group C). Every group has12mice. On day0, immune group A and group B with PTg and CFA, on day14immune the two groups with PTg and IFA. In addition,on day0、7、14、21the group A receive BMSCs treatment through tail vein injection. BMSCs come from homologous C57BL/6mice, after separation、culture、identify, harvest the P3BMSCs. On28day of the experiment, pick eye ball and collect the blood, detect the serum levels of TT3、TT4、TgAb、TPOAb、TMAb、IFN-γ、IL-17of the three groups. All the mice were sacrificed, separate and stain the thyroid with hematoxylin-eosin. Lymphocytes infiltration degrees of thyroid were examined.Results:l.The serum levels ofTgAb、TPOAb、TMAb and degrees of lymphocytes infiltration of thyroid in group A were higher than group C, there were statistics significant (P<0.05); the serum levels of TgAb、TPOAb、TMAb and lymphocytes infiltration of thyroid in group B were higher than group C, there were statistics significant (P<0.05); TT4in groupA、Bwas lower than group C(P<0.05); TT3between the group A、B、C have no statistical significance(P>0.05).2. The serum levels ofTgAb、TPOAb、TMAb and the degree of lymphocytes infiltration of thyroid in group B were higher than group A,TT4was lower than groupA, there were statistics significant (P<0.05).3. The serum levels of IFN-γ、IL-17in the group A、B、C have statistics significance (P<0.05);The IFN-γ、IL-17levels in group B were higher than group A, they have statistics significance (P<0.05); The IFN-γ、IL-17levels in group A were higher than group C, they have statistics significance (P<0.05); The IFN-γ、IL-17levels in group B were higher than group C, statistics significance exists (P<0.05).4. In group A there exist positive correlation between the serum level of IFN-y and the TPOAb、IL-17and TPOAb, the correlation has statistics significance (P<0.05); in group B there exist positive correlation between the level of IFN-γ and TPOAb、IL-17and TPOAb, the correlation has statistics significance (P<0.05).Conclusions:1. Group A、B were immune subcutaneously twice by pTg with adjuvant, serum levels ofTgAb、TPOAb、TMAb were higher than normal control group, TT4was lower than group C after the experiment, and the thyroid exhibited lymphocytes infiltration, have chief characters of EAT, the immune induction was successful.2. The antithyroid autoantibodies-TgAb、TPOAb、TMAb and pro-inflammation cytokines-IFN-γ、IL-17, lymphocytes infiltration in group A lower than group B, TT4higher than group B, means that homologous BMSCs could protect against immune damage, have therapy effect for EAT. The mechanism need further exploring.3. Serum levels IFN-γ、IL-17cytokines were higher in A、B than group C, IFN-y as chief cytokines of Thl cells, and IL-17as chief cytokines of Thl7cells, both were pro-inflammation cytokines, may involve in the pathogenesis of EAT. TPOAb as one of the titers of EAT, is the special immune markerof EAT, the positive colleration between IFN-γ、IL-17and TPOAb, exhibits the relationship of IFN-γ、IL-17and TPOAb,we conclude that both Th1、Th17involved in the pathogenesis of EAT.
