Objective:The model of Endometriosis(EMT) in Wistar rat was established by self-endometrium transplantation. The effect of Compound Curcuma Decoction on the EMT model rats were investigated on morphology in eutopic and ectopic endometrial tissue and cluster of differentiation antigen3,4,8(CD3, CD4and CD8) in the peripheral blood of EMT model rats.Meanwhile, the role of Compound Curcuma Decoction regulating endometriosis helper T cells1and helper T cells2ratio (Thl/Th2) balance in rats were also investigated. Explore the therapeutic mechanism of Compound Curcuma Decoction from the perspective of immunomodulatory and to provide more experimental evidence for the clinical application of Compound Curcuma decoction.Methods:The models of Endometriosis(EMT) in70healthy female Wistar rats were established by self-endometrium transplantation without testing estrous cycle.4weeks after transplantation, we made the second laparotomy to select right rats according to the inclusion and exclusion criteria. And then randomly assigned into4group:the high-dose of CCD,low-dose of CCD,gestrinone group and model control group.Meanwhile,we select10rats as the normal control group.After4weeks’ treatment, we made the third laparotomy to check the bulk and weight of transplant,to observe the pathomorphological changes with light microscopes.Using flow cytometry to detect endometriosis CD4/CD8and Thl/Th2of peripheral blood in rats. Experimental results are analyzed by SPSS17.0statistical software,the comparisons between two with independent samples t-test,the comparisons among multiple groups are processed by completely random variance analysis,the comparisons between mean of two groups are detected by Students-Newman-Keuls(SNK) method.The condition of P<0.05is considered to have statistical significance.Results:1、The second laparotomy (4weeks after modeling) confirmed:The endometrial implants in the rats became cystic structures that were composed of endometrium;endometrial gland and stroma.The macroscopic and histological appearance were consistent with endometriosis.Under the method of establishing the model of EMT by self-endometrium transplantation without testing estrous cycle,the success rate was80%.No significant difference was found in the volumes of grafts between model control group and each CCD therapy group on the second laparotomy(P>0.05).Treated for4weeks,the volume of each CCD therapy group on the third laparotomy was differently reduced compared with the model control group(P<0.05). No significant difference was found in the weight of grafts between each CCD therapy group and gestrinone group(P>0.05).2、From the histological point of view,ectopic endometrial tissues show different degrees of atrophy in treatment groups compared with the model control group,with the number of glands becoming less and the area becoming narrow,covered epithelial becoming thin,meanwhile epithelial cells show low columnar or flat or even loosely arranged and interstitial cells was becoming less, the number of blood vessels have diminished.3、The peripheral blood CD3, CD4of EMT model rat was significantly lower than the normal control group (P<0.05),CD4/CD8decreased compared with the normal control group (P<0.05),CD8on a downward trend, but the difference was not statistically significant.Each treatment group compared with model group,CD3、 CD4and CD4/CD8increased, there is a statistically significant difference (P<0.05).CCD group and gestrinone group, CD3, CD4and CD4/CD8were no significant differences.4、EMT model rats compared with the normal control group, the CD4+IL-4+of peripheral blood increased (P<0.05), Thl/Th2decreased (P <0.05), CD4+IFN-gamma+has no significant change.Compound Curcuma decoction compared with the model group, CD4+IL-4decreased, Thl/Th2rise, the difference was statistically significant (P<0.05).There was no significant difference in CD4IL-4、Th1/Th2between CCD group and gestrinone group.Conclusion:1、CCD may inhibits the development of endometriosis and reduce the size and weight of grafts,reduce abdominal adhesions, improve the pathological situations of ectopic endometrial tissue, the effect of HCCD group is most obvious.2、 The existence of immune dysfunction in EMT model rats was confirmed,CDS, CD4, CD8, CD4/CD8in peripheral blood decreased, the abnormal state of T lymphocyte subsets in EMT model rats can be adjusted by CCD,the effect of HCCD group is most obvious.3、CD4+IL-4+rise Thl/Th2decreased, CD4+IFN-gamma+has no significant change in the peripheral blood of EMT model rats, Th1/Th2imbalance in EMT model rats can be adjusted by CCD.
