In this study, the marine bioactive strains were screened with dualbiological activity model, and metabolites from the bioactive strainsENP702#were isolated by the directed activity screening technology,which was established using High Performance Liquid Chromatography(HPLC)-Bioactivity Fingerprint Profiles (BFP)-based platforms in a fixedtime. The following major conclusions can be drawn from the study:(1) Eight marine microorganism which were separated from EastChina Sea were screened by dual bioactivity model of antioxidantive(DPPH method) and antitumor activities (MTT method). Two strainsshowed strong antioxidantive, and their inhibiting rate were over90%atthe concentration of2mg·mL-1. In the antitumor activity test, theinhibition activity against HeLa and H460cells were more than50%atthe concentration of50μg·mL-1.(2) The marine active strain ENP702#was cultured in large-scale. Meanwhile, it was analyzed by ITS sequence, combined withmorphological character and physiological and biochemical property,the strain was identified as Penicillium sp. ENP702.(3) The mycelium and culture filtrate extracts of ENP702#wasanalyzed by the HPLC-BFP, and the bioactivity was evaluated by themodel of trypsin inhibitor activity and antioxidantive activity. Thedirected recognition of bioactive metabolites from ENP702#wasestablished by the HPLC-BFC method.(4) Seven bioactive metabolites from ENP702#was isolated usingflash preparative chromatography according to the directed activityrecognition of HPLC-BFC. They are Mannitol (1), Erythritol (2),5,7,22-triene-3β-hydroxy-ergost-erol (3), Allantoin (4),6,7,8-trimethoxy-coumarin (5),1-O-β-D-glucopyranosyl-(2S,3S,4E,9E)-2-(2’R,3’E)-2’-hy-drox-yoctadec-3’-enoylamino)-10-methyl-4,9-dien-1,3-diol (6), cyclo-(I-le-Val)(7). At the concentration of100μg·mL-1, allantoin showed goodDPPH free radical scavenging activity with the inhibition rate of42.59%,coumarin showed the significative inhibition rates of36.42%againsttrypsin.
