ObjectiveTo detect the expression of Dicer in hilar cholangiocarcinoma tissue and malignantcholangiocyte(QBC939).To investigate relationship between Dicer expression andclinicopathological characteristics and prognosis after radical surgery resection of hilarcholangiocarcinoma.Methods1. The expression of Dicer in40specimens of hilar cholangiocarcinoma and10specimens of normal bile duct tissues was detected using immunohistochemistry.2. Western blotting and RT-PCR were used to investigate the expression of Dicer inhuman common bile duct adenocarcinoma cell line QBC939and human intrahepaticbiliary epithelial cell line HIBEpic.3. The relationship between Dicer expression and clinicopathological characteristics wasanalyzed. A Kaplan–Maier analysis was performed to analyze disease-free survival(DFS) and overall survival (OS) after radical surgery resection of hilarcholangiocarcinoma.ResultsCompared to normal bile duct tissues, Dicer was significantly down-regulated in hilar cholangiocarcinoma tissues (p<0.05). The expression of Dicer in human common bileduct adenocarcinoma cell line QBC939and human intrahepatic biliary epithelial cell lineHIBEpic was detected respectively in protein and mRNA level, which showed lowerexpression in QBC939than in HIBEpic (p<0.05).The expression of Dicer in tissue andcell line are both significantly down-regulated. Furthermore, the expression of Dicer inwell differentiation adenocarcinoma is higher than poor and moderate differentiationadenocarcinoma. The univariate statistics shows the low expression of Dicer protein wassignificantly correlated with shorter disease-free survival and overall survival of hilarcholangiocarcinoma patients after radical surgery resection (p<0.01). Multivariateanalysis revealed that the expression of Dicer was the most important predict factor forprognosis after radical surgery resection of Hilar cholangiocarcinoma (p<0.05).ConclusionsThe expression of Dicer both in hilar cholangiocarcinoma tissue and cell line issignificantly down-regulated. The level of Dicer is correlated with differentiation degree ofhilar cholangiocarcinoma. Low expression of Dicer predicts poor prognosis in hilarcholangiocarcinoma. Therefore, we hypothesise that the expression levels of Dicer couldbe used as a potential prognostic marker for hilar cholangiocarcinoma.
