The Expression And Clinical Significance Of PIWIL2in Hilar Cholangiocarcinoma And Malignant Cholangiocyte

Objective:To investigate the expression of PIWIL2in hilar cholangiocarcinoma and （QBC939）malignant cholangiocyte. And analysis the relationship between PIWIL2andclinicopathological characteristics and prognosis of radical resection in hilarcholangiocarcinoma.Methods:1. Immunohistochemistry was used to detect the expression of PIWIL2in41specimens ofhilar cholangiocarcinoma and10specimens of normal bile duct tissue.2.Western-blot and immunocytofluorescent were used to detect the expression of PIWIL2in cholangiocarcinoma cell lines QBC939and bile duct epithelial cell lines HIBEpic.3.Survival use the Kaplan-Meier method to univariate and multivariate analysis thesurvival time of hilar cholangiocarcinoma patients who undergoing radical surgery;meanwhile analysis the relationship between PIWIL2expression and clinicopathologicalcharacteristics.ResultThe expression of PIWIL2in hilar cholangiocarcinoma tissues and QBC939cholangiocarcinoma cell lines was significantly higher than in normal bile duct tissues andHIBEpic bile duct epithelial cell lines; The expression of PIWIL2in poor and moderatedifferentiation cholangiocarcinoma adenocarcinoma was significantly higher than welladenocarcinoma. Univariate analysis showed that the high expression of PIWIL2withshort-survival time after radical surgery resection. Multivariate analysis showed that the expression level of PIWIL2was a independent prognostic factor for estimate after radicalsurgery resection of cholangiocarcinoma.ConclusionsThe expression of PIWIL2is upregulated in hilar cholangiocarcinoma tissue andcholangiocarcinoma cell line.The level of PIWIL2expression is associated with TNM fortumor stage and degree of differentiation;The expression of PIWIL2is a independentfactor for prognosis after radical surgery resection of hilar cholangiocarcinoma.
