Objectives To detect and compare the expressions of HIF-1α and LOX in NSCLC;To analyze the relationship of the two genes expressions in clinical and pathologicalcharacteristics; This study will be useful for understand the mechanism of lymphnodemetastasis and invasion in NSCLC and for screening tumor related markers, designingdetect methods to evaluate the potential of tumor metastasis and invasion, finding newtheory and therapeutic target for effective regimen ofinterrupting metastasis andinvasion of NSCLC.Methods35cases of NSCLC were selected randomly from Department ofThoracicSurgery of Tongji Hospital between October2010and February2011. Byreal-time fluorescence quantitative polymerase chain reaction andimmunohistochemistry detecting HIF-1α mRNA, LOX mRNA and theircorresponding proteins in TT and SNT. Statistical analysis was carried out bycompiling the expressions of two genes with clinical and pathological information ofthe patients, and then got the correlation between each indicator and the meaning ofthem.Results1. The expression of HIF-1α mRNAand LOX mRNAin NSCLCHIF-1α and LOX expression at the mRNA level was investigated in35NSCLCby FQ-PCR.22out of35NSCLC showed higher HIF-1α mRNA expression in theirTT than SNT. In TT, the median HIF-1α mRNA levels is1.107×10-3. In SNT, the median HIF-1α mRNA levels is0.975×10-3. No significant association was foundbetween them.27out of35NSCLC show higher LOX mRNA expression in their TTthan SNT. In TT, the median LOX mRNA levels is8.025×10-3. In SNT, the medianLOX mRNA levels is3.716×10-3, the LOX mRNA levels in TT were significanthigher than those observed in SNT(P<0.01). High HIF-1α mRNA levels showedsignificant correlations with T stage(P=0.004). High LOX mRNA levels showedsignificant correlations with gender(P=0.034)、smoking status(P=0.034)、p-TNMstage (P=0.028)and T stage(P=0.008).2. The expression of HIF-1α and LOX in NSCLCHIF-1α and LOX expression at the protein level was investigated in35NSCLCby IHC.14out of35NSCLC showed HIF-1α high expression,18out of35NSCLCshowed LOX high expression. No significant associations were found betweenHIF-1α protein and clinicopathologic features. High LOX protein expression showedsignificant correlations with Ⅱ+Ⅲ stage(P=0.012)、the presence of lymph nodemetastasis(P=0.044) and T2-T4stage(P=0.001).3. The relationship between HIF-1α and LOXFQ-PCR using two random variables correlation analysis, r=0.414, p<0.05, IHCdata using paired chi-square and calculate the correlation coefficient, r=0.443,p<0.01,statistical analysis showed that: HIF-1α and LOX have significant positive correlationin NSCLC.Conclusions1. The high expression of HIF-1α mRNA, LOX mRNAand their correspondingproteins in NSCLC may play an important role in the promotion of NSCLC.2. The high expression of HIF-1α mRNA, LOX mRNAand their correspondingproteins showed significant correlations with p-TNM stage、the presence of lymphnode metastasis and T stage, which may be correlated with the invasion andmetastasis of NSCLC.
